摘    要:目的:通过观察填精通络方及其拆方对糖尿病性勃起功能障碍大鼠性激素及阴茎海绵体eNOS酶表达的影响,探讨填精通络方在治疗糖尿病性勃起功能障碍的作用机制。方法:将60只SPF级雄性SD大鼠随机分为正常组、模型组、对照组、填精组、通络组及填精通络组6组,每组10只。除正常组外,其余组以腹腔注射链脲佐菌素制备糖尿病大鼠模型,任意时间血糖>16.67 mmol·L-1为糖尿病成模标准,然后用阿朴吗啡阴茎勃起实验筛选糖尿病性勃起功能障碍大鼠模型。成模后,对照组予以0.2 mg·kg-1他达拉非灌胃,每周两次;填精通络组予以填精通络方以9.3 g·kg-1·d-1灌胃,填精组予以拆方Ⅰ号以7.2 g·kg-1·d-1灌胃,通络组予以拆方Ⅱ号以2.1 g·kg-1·d-1灌胃,正常组与模型组分别灌服等体积蒸馏水,连续30天。取腹主动脉血清,用酶联免疫分析法测定各组大鼠的性激素水平的差异;便携式血糖仪检测各组大鼠的血糖水平;免疫荧光检测离体阴茎组织eNOS 酶含量。结果:与模型组比较,填精组、通络组及填精通络组的血糖明显降低(P < 0.01),阴茎海绵体eNOS酶的表达改善,血清睾酮(T)含量明显上升(P < 0.05),而卵泡刺激素(FSH)、黄体生成素(LH)含量变化不明显(P > 0.05);与对照组比较,填精通络组的血糖明显降低(P < 0.01),阴茎海绵体eNOS酶的表达改善,血清T含量明显上升(P < 0.05),而血清FSH、LH含量变化不明显(P > 0.05);与通络组比较,填精组和填精通络组的血糖明显降低(P < 0.05),但填精组阴茎海绵体eNOS酶的表达改善不如通络组,而填精通络组阴茎海绵体eNOS酶的表达改善优于通络组。结论:填精通络方能明显降低大鼠血糖水平,改善大鼠性激素水平和阴茎海绵体eNOS酶表达,从而改善大鼠的勃起功能障碍。

Abstract:Objective: By observing the effects of Tianjing Tongluo decoction and its disassembled formulae on sex hormones and the expression of penile cavernosa eNOS enzyme in diabetic rats with erectile dysfunction, to investigatethe mechanism of Tianjing Tongluo decoction in the treatment of diabetic erectile dysfunction. Methods: A total of 60 SPFmale SD rats were randomly divided into 6 groups: normal group, model group, control group, Tianjing group, Tongluogroup and Tianjing Tongluo group, with 10 in each group. In addition to the normal group, other groups prepared diabeticrat models with intraperitoneal injection of streptozotocin. Blood glucose more than 16.67 mmol·L-1 at any time was thestandard for diabetic modeling, and then apomorphine penile erectile experiment was used to screen the diabetic erectiledysfunction rat model. After the model was established, the control group was given 0.2 mg·kg-1 tadalafil gavage twice aweek. The Tianjing Tongluo group was given Tianjing Tongluo Decoction with 9.3 (g·kg-1)·d-1. The Tianjing group wasgiven the disassembly 1 with 7.2 (g·kg-1)·d-1. The Tongluo group was given the disassembly 2 with 2.1 (g·kg-1)·d-1. Themodel group and normal group respectively were received irrigation clothing such as distilled water volume, 30consecutive days. The serum of abdominal aorta was taken and the difference of sex hormone levels in each group wasdetermined by enzyme-linked immunoassay. Blood glucose levels were measured by portable glucose meter in eachgroup. The content of eNOS enzyme in isolated penile tissue was detected by immunofluorescence. Results: Compared with the model group, the blood glucose of the Tianjing group, the Tongluo group and the Tianjing Tongluo group wassignificantly decreased (P < 0.01). The expression of penile spongiform eNOS enzyme was improved, and the serumtestosterone level was significantly increased (P < 0.05), while the serum FSH and LH contents were not significantlychanged (P > 0.05). Compared with the control group, the blood glucose of the group was significantly decreased (P <0.01). The expression of penile spongiform eNOS enzyme was improved, and the serum testosterone level wassignificantly increased (P < 0.05), while the serum FSH and LH contents were not significantly changed (P > 0.05).Compared with the Tongluo group, the blood glucose of the Tianjing group and the Tianjing Tongluo group wassignificantly decreased (P < 0.05), but the expression of penile spongiform eNOS enzyme was improved in the Tianjinggroup was not as good as that in the Tongluo group, and the expression of penile spongiform eNOS enzyme was improvedin the Tianjing Tongluo group was better than that in the Tongluo group. Conclusion: Tianjing Tongluo Decoction cansignificantly decrease the blood glucose level of rats, improve the level of sex hormones and the expression of penilespongiform eNOS enzyme was improved, and improve the erectile dysfunction of rats.
