| 摘 要： | 目的研究药物超声灸疗纳米乳凝胶剂的制备工艺。方法以复方辣椒碱乳膏(辣椒碱、薄荷脑、樟脑、冬青油)为模型药物,HLB法制备纳米乳,再制备复方辣椒碱纳米乳凝胶。采用伪三元相图对纳米乳中的表面活性剂与助表面活性剂进行优选;用正交设计对凝胶剂中的透皮促渗剂进行优选;采用Franz扩散池,进行体外经皮渗透考察;并对超声作用条件进行考察。结果以处方药物与氮酮的混合物为纳米乳油相,吐温80为表面活性剂,1,2-丙二醇为助表面活性剂(KM=3∶1),纳米乳平均粒径17.2 nm;透皮促渗剂为:1%氮酮+20%丙二醇+20%甘油+1%PEG400;超声最优条件为1.5 W(305 kHz),作用时间为3 min。体外透皮释放曲线显示,超声作用纳米乳凝胶前2 h累积渗透量为无超声作用下的2倍以上,24 h的累积渗透量约为无超声作用下的3.5倍。结论体外试验显示,制备的复方辣椒碱纳米乳凝胶在超声波的辅助作用下,释药快而持久,兼具速效与缓释的双重作用,优于普通乳剂。
