枸杞叶新资源药材的生药学研究

摘    要:目的 对枸杞叶生药学进行研究,为其新资源药材质量标准的建立提供参考。方法 分别采用形态组织学法和薄层色谱法研究枸杞叶性状、显微及薄层色谱特征,并采用高效液相色谱法(HPLC)对枸杞叶中的甜菜碱进行定量分析。结果 系统描述了枸杞叶药材性状和显微特征,主要包括枸杞干燥叶颜色、形状、大小及叶端、叶柄等性状特征。观察到叶柄密布棒槌状小腺毛、主脉下表面圆齿状等微性状特征;叶横切面显微图完整且详细地呈现了下陷气孔、腺毛、砂晶囊、簇晶、方晶、栅栏组织、厚角组织等重要特征;叶表面有不定式气孔和不等式气孔类型,观察到保卫细胞的形状和大小等特征;得到枸杞叶中芦丁和绿原酸的薄层色谱图;20批宁夏枸杞叶中甜菜碱的平均质量分数为4.856%。结论 在《宁夏中药材标准》《宁夏中药饮片炮制规范》中宁夏枸杞叶标准的基础上,进一步丰富了其性状和显微特征的描述,首次对其微性状特征进行了描述,建立了枸杞叶薄层色谱鉴别和HPLC含量测定方法。

Abstract:Objective To carry out pharmacopharmsitic study of Lycii Folium and thereby to lay a theoretical basis for developing the quality standard of this medicinal from new resources.Methods The morphological observation, microscopy, and thin-layer chromatography (TLC) were employed to identity the characteristics of Lycii Folium, and betaine was quantified by high performance liquid chromatography (HPLC).Results The morphological and microscopic characteristics of Lycii Folium, mainly including the color, shape, size, leaf apex, and petiole, were described. The petiole was densely covered with wooden club-shaped glandular trichomes, and the cuticle of the lower epidermis of the midrib was scalloped. The micrograph of the leaf cross-section presented important features such as sunken stomata, glandular trichomes, sand crystal sacs, clustered crystals, cubic crystals, palisade structure, and collenchyma. There were anomocytic type stomas and anisocytic type stomas on the leaf surface, and the shape and size of guard cells were also clarified. A TLC for the identification of rutin and chlorogenic acid in Lycii Folium was established. The average content of betaine in 20 batches of Lycium barbarum leaves was 4.856%.Conclusion This paper added the traits and microscopic characteristics of L. barbarum leaf in related standards in Ningxia Traditional Chinese Medicine Standard and Preparation Specification of Ningxia Traditional Chinese Medicine, described the microscopic traits for the first time, and developed the TLC or identification and HPLC for content determination of Lycii Folium.
