鼻脑通鼻用喷雾剂对实验性偏头痛动物ET-1、CGRP含量的影响

摘    要:目的:研究鼻脑通鼻用喷雾剂对实验性偏头痛小鼠内皮素-1(ET -1)、降钙素基因相关肽(CGRP)含量的影响。方法:75只ICR小鼠,雌雄各半,随机分为5组,即对照组、模型组、鼻脑通鼻用喷雾剂低、中、高剂量组,每组15只。采用硝酸甘油皮下注射制备小鼠实验性偏头痛模型,造模后20min开始记录小鼠前肢搔头的次数,记录10min;造模4h后腹主动脉取血,测定血浆中ET -1、CGRP指标。结果:与对照组相比,模型组动物搔头次数显著增多(P〈0.001);提前给予鼻脑通鼻用喷雾剂3个剂量后,造模小鼠的搔头次数均比模型组显著减少(P〈0.05,P〈0.01,P〈0.001)。与对照组相比,模型组动物血浆ET-1含量显著升高(P〈0.001)、CGRP含量升高(P〈0.01);而与模型组相比,提前给予鼻脑通鼻用喷雾剂3个剂量后,造模的动物血浆中、高剂量组ET-1含量显著降低(P〈0.001,P〈0.001),低、中、高剂量组CGRP含量均降低(P〈0.05,P〈0.01,P〈0.叭)。结论:鼻脑通鼻用喷雾剂能减少实验性偏头痛动物的搔头次数,其治疗偏头痛的机制可能与降低动物血浆ET-1、CGRP含量有关。

Abstract:Objective: To study the effects of binaotong nasal spray on ET-1 and CGRP levels in experimental mice with migraine. Methods: 75 ICR mice were randomly divided into 5 groups( n = 15): control group, model group, binaotong nasal spray groups of low dose, middle dose and high dose. Experimental model of migraine mice was performed by hypodermic injection of nitroglycerin. 20 min later, the frequency of scratching head was record for 10 min. The plasma ET-1, CGRP levels were detected respectively 4 h later. Results: The frequency of scratching head in model group was increased significantly compared with control group ( P 〈 0. 001 ) . However, treatment of binaotong nasal spray at three concentrations reduced the frequency of scratching head ( P 〈 0. 05, P 〈 0. 01, P 〈 0. 001 ) . In nitroglycerin-induced migraine model mice, plasma ET-1 level was increased significantly ( P 〈 0. 001 ) and plasma CGRP level was increased( P 〈 0. 01 ) compared with control group. However, plasma ET-1 levels were decreased markedly in binaotong nasal spray-treated groups of middle dose and high dose (P 〈 O. 001, P 〈 O. 001 ) plasma CGRP levels were decreased( P 〈 0.05, P 〈 O. 01, P 〈 0. 01 ) . Conclusion: Binaotong nasal spray could decrease the frequency of scratching head in experimental migraine mice. Furthermore, its mechanisms may be decreaseing plasma ET-1 and CGRP level
