滋阴活血方预处理对大鼠缺血再灌注心肌保护作用研究

摘    要:目的研究滋阴活血方预处理对大鼠心肌缺血再灌注损伤的保护作用。方法使用wistar大鼠作为实验动物,分别设正常对照组、模型组、滋阴活血方(高、中、低)三个剂量组、阳性药对照组、复方丹参滴丸组。动物于造模前2天给药,第三天造模;再灌注2小时后,腹主动脉取血,测定血清LDH、CK、CK—MB、MDA、SOD含量,观察心肌梗塞程度。结果①滋阴活血方高、中两个剂量组梗塞程度明显减轻,梗塞区重占全心脏及左心室的百分比与模型组比较均有显著性差异。②模型组动物血清LDH、CK及CK—MB含量均明显增加;滋阴活血方各剂量组无明显影响。③滋阴活血方高、中两个剂量组SOD值明显增加,与模型组比较有显著性差异(P〈0.01)。结论①滋阴活血方预处理能够防止或减轻心肌缺血再灌注损伤。②滋阴活血方对缺血再灌注损伤的保护作用可能是通过预处理的途径产生的。③滋阴药物预处理对缺血再灌注损伤的保护作用符合扶正祛邪的中医传统理论。

Abstract:Objective To observe the effects of the Ziyinhuoxuefang (ZF) in protection of myocardial ischemia reperfusion injury (MIRI) in an enhancing ischemic preconditioning (IPC) model of the rats. Methods The male Wistar albino rats were divided in seven groups, control group, model group, herbal group,ZF group( H,M ,L, three dosage group) and Isosorbide Dinitrate group( ID group). All rats drenching by medicine for two days before operation. The ischemica-reperfusion model is made by the third day. Observe the effects of infarct size, and LDH,CK,CK-MB,SOD、MDA. Results ① Determination of Infarct SizeIn the reperfusion group, the weight of heart and left ventricle was not significance in any case. In the model group, the ratios of the infarct size of the area at heart and left ventricle were (33.18 ± 4. 87)% and(42. 54 ±6. 83)% ; ZF (H,M) groups were considered significant raised than model group; the same as the ID group. ( H ) group was the best one. ② The contents of serum LDH,CK and CK-MB in all groups above were significantly higher than that of the sham operated (P 〈0. 001 ). Compared to that of the administration groups was not significance in any case. ③ The SOD and MDA production in serum of model group was lower than sham operated group;ZF (H,M) groups were considered significant raised than model group. All administration groups Malonyldialdehyde production was not significance in any case. Conclusions ① Effect of ZF on Myocardial Ischemia Reperfusion Injury. ② Effect of ZF on the Mechanism in Protection of MIRI. ③ Fuzhengquxie is Based on the Basic Theory of Traditional Chinese Medicine.
