Authentication of Lysimachia christinae and its adulterants by analysis of ITS2 sequences

Abstract:Objective To identify Lysimachia christinae and its adulterants,and to secure the quality and safety in medication of this medical material.Methods The PCR technique was performed to get the ITS2 regions and purified PCR products were sequenced.Sequence assembly and consensus sequence generation were performed using the CodonCode Aligner.Sequence analyses were carried out by MEGA4.The phylogenetic tree was constructed using the neighbor-joining and Kimura 2-parameter methods.Moreover,The ITS2 secondary structure was predicted using the ITS2 database and website found by Koetschan et al.Results A significant difference was observed in the ITS2 sequences between Lysimachia christinae and its adulterants.NJ tree that is based on ITS2 sequences and secondary structure of ITS2 can distinguish between Lysimachia christinae and its adulterants.Conclusion ITS2 can serve as a valuable marker for identifying Lysimachia christinae and its adulterants,and it has a broad application in the identification of traditional Chinese medicine.
