燧心胶囊对慢性心力衰竭大鼠凋亡相关基因Bcl-2和Bax表达的影响

摘    要:目的:以慢性心力衰竭大鼠为研究对象,测定心肌组织Bcl-2、Bax mRNA水平及其蛋白表达,以探究燧心胶囊干预心室重塑的作用机制。方法:将60只大鼠随机分为假手术组、模型对照组、地高辛组、卡托普利组、燧心胶囊低剂量组、燧心胶囊高剂量组。采用腹主动脉缩窄术建立大鼠慢性心力衰竭模型,连续给药12周。Real time PCR检测心肌细胞凋亡基因Bcl-2 mRNA、Bax mRNA水平;免疫组化法观察大鼠心肌细胞凋亡蛋白Bcl-2、Bax的表达。结果:与模型对照组比较,卡托普利组、燧心胶囊低、高剂量组大鼠Bcl-2 mRNA水平及其蛋白表达增高,Bax mRNA水平及其蛋白表达降低,Bcl-2/Bax则显著性增高,差异具有统计学意义(P0.05或P0.01)。其中燧心胶囊高剂量组对Bcl-2和Bax mRNA及其蛋白的调控程度较低剂量组显著(P0.05)。结论:高剂量燧心胶囊能够调控凋亡基因Bcl-2和Bax mRNA水平及其蛋白表达,抑制心肌细胞程序性死亡,延缓心力衰竭的进程。

Abstract:Objective:To research the mechanisms of Suixin capsule interfering in chronic heart failure(CHF)rats,we measured Bax and Bcl-2 mRNA levels and protein expressions,in order to lay the foundation for further clinical application.Methods:Partially banding abdominal aortic artery to achieve CHF rats model,60 rats were randomly divided into six groups:sham group,control group,digoxin group,captopril group,low-dosage group and high-dosage group.Real time PCR was used to observe apoptosis-related gene Bax and Bcl-2 mRNA levels;immunohistochemistry was used to detect the expressions of Bax and Bcl-2 in rat myocardial cell.Results:Compared with control group,Bax levels in captopril group and high-dosage group reduced,whereas Bcl-2 level increased,and Bcl-2/Bax increased(P<0.05 or P<0.01).The regulation of Bax and Bcl-2 in high-dosage group was more significant than that in low-dosage group(P<0.05).Conclusions:High dose of Suixin capsule can regulate apoptosis-related gene Bax and Bcl-2,and inhibit the apoptosis of cardiocytes.Therefore it can transform the failure heart from decompensation to compensation and postpone the progress of heart failure.
