补肾活血方对不明原因复发性流产患者分泌晚期血清中IL-11、IL-11R的影响研究

目的探究补肾活血方对肾虚血瘀型不明原因复发性流产(URSA)患者分泌晚期血清中IL-11及其受体的影响。方法第一部分实验测定肾虚血瘀型URSA患者15例(6~10周)及无自然流产病史正常早孕的15例(6~10周)流产蜕膜组织中IL-11、IL-11R及蜕膜化标志物PRL、IGFBP-1mRNA的含量。第二部分实验将未孕的肾虚血瘀型URSA患者80例,随机分为服用中药补肾活血方实验组(40例)、服用西药阿司匹林对照组(40例),收集有过至少一次妊娠史的正常妇女40例作为正常组,检测未孕的肾虚血瘀型URSA患者治疗前后与正常妊娠妇女分泌晚期血清中IL-11、IL-11R及蜕膜化标志物PRL、IGFBP-1的表达水平。结果IL-11、IL-11R、PRL及IGFBP-1mRNA表达在URSA组中较正常组中低;IL-11、IL-11R、PRL、IGFBP-1水平在URSA患者血清中较正常妊娠女性血清中表达低;URSA患者口服补肾活血方治疗4周后,血清中IL-11、IL-11R、PRL、IGFBP-1表达升高,结果好于阿司匹林,且与正常人血清无显著差异。结论肾虚血瘀型URSA患者蜕膜组织中IL-11、IL-11R及蜕膜化标志物PRL、IGFBP-1表达减少,与URSA妊娠早期蜕膜化不足流产相关。补肾活血方可能通过适当升高肾虚血瘀型URSA患者分泌晚期血清中IL-11、IL-11R及蜕膜化标志物PRL、IGFBP-1的表达,促进滋养细胞蜕膜化和胚胎发育,并且比西药阿司匹林效果更显著。

Effect of Bushen Huoxue Recipe on IL-11 and IL-11R in Later Period Serum of Patients with URSA

Objective To investigate the effect of Bushen Huoxue Recipe on the secretion of IL-11 and its receptors in the serum of patients with URSA of kidney deficiency and blood stasis type.Methods The first part of the experiment was to determine the levels of IL-11,IL-11 R,PRL and IGFBP-1 mRNA in 15 cases(6~10 weeks)of URSA of kidney deficiency and blood stasis type and 15 cases(6~10 weeks)of normal early pregnancy without spontaneous abortion.In the second part of the experiment,80 patients with non-pregnancy URSA of kidney deficiency and blood stasis type were randomly divided into the experimental group of Chinese medicine Bushen Huoxue Decoction(40 cases)and the western medicine aspirin control group(40 cases).Forty normal women were used as the normal group.The expression levels of IL-11,IL-11 R,PRL and IGFBP-1 in later period serum were detected before and after treatment in non-pregnant kidney deficiency and blood stasis type URSA patients.Results Expressions of IL-11,IL-11 R,PRL and IGFBP-1 mRNA were lower in the URSA group than those in the normal group.IL-11,IL-11 R,PRLand IGFBP-1 receptor levels were more normal in URSA patients.The expression of serum in women was low.The expressions of IL-11,IL-11 R,PRL and IGFBP-1 in serum of patients with URSA were increased after oral administration of Bushen Huoxue Recipe for 4 weeks.The results were better than aspirin and there was no significant difference compared with normal human serum.Conclusion The expressions of IL-11,IL-11 R,decidualization markers PRL and IGFBP-1 in decidual tissue of patients with URSA of kidneydeficiency and blood stasis type are decreased,which is associated with dysplasia of URSA in early pregnancy.Bushen Huoxue Recipe may promote the expressions of IL-11 and IL-11 R,decidualization markers PRL and IGFBP-1 in later period serum to promote decidualization and embryonic development of trophoblasts,and it is more effective than Western medicine aspirin.
