白藜芦醇抗肝癌Bel-7402及调节荷瘤鼠IL-8分泌机制的研究

摘    要:目的:探讨白藜芦醇离体与在体对肝癌细胞Bel-7402的抑制作用及可能的效应机制。方法:MTT比色法测定Bel-7402细胞生长抑制率,Western blotting测定Bel-7402细胞Bcl-2水平,流式细胞术分析细胞周期,流式细胞染色检测细胞内IL-8,半定量RT-PCR与双抗体夹心ELISA法检测荷瘤小鼠IL-8 mRNA表达与含量。结果:白藜芦醇可以时间-剂量依赖性抑制Bel-7402细胞增殖,诱导凋亡,影响细胞周期分布。此外,白藜芦醇还能剂量依赖性抑制荷瘤小鼠细胞内IL-8产生及其mRNA与蛋白含量。结论:白藜芦醇离体与在体均能发挥有效的抗肝癌生物学活性和免疫调节作用。

Abstract:OBJECTIVE: To explore the anti-tumor effect of resveratrol on the hepatocellular carcinoma cell line Bel-7402 both in vitro and in vivo. METHODS: MTT colorimetry was used to measure the tumor cell growth repression rate. The level of Bcl-2 was measured by Western blotting. The cell cycle of Bel-7402 and intracellular IL-8 were analyzed by flow cytometry. The expression of IL-8 mRNA was determined by semiquantitative RT-PCR and quantification assay of IL-8 was determined by ELISA. RESULTS: Our findings indicated that resveratrol inhibited proliferation, induced apoptosis, and influenced cell cycle of Bel-7402 cells at a dose-and time-dependent manner in vitro. Furthermore, resveratrol could exert a dose-related down-regulatory effect on IL-8 in Bel-7402 bearing mice. CONCLUSION: It suggestes resveratrol might have chemotherapeutic potential and immunomodulation action to Bel-7402 to cellular carcinoma both in vitro and in vivo.
