中药材苦地胆的DNA指纹鉴定

摘    要:采用分子生物技术包括任意引物聚合酶链式反应(AP-PCR)和随意引物扩增的多态性DNA(RAPD)方法扩增菊科植物地胆草、白花地胆草和假地胆草以及商品药材苦地胆的基因组DNA,获得清晰可靠的DNA指纹图谱。根据琼脂糖凝胶上显示的DNA带型差异可鉴别苦地胆和其混淆品,同时,根据DNA指纹图谱的相似度指数值计算,证实福建、台湾、香港、澳门商品药材苦地胆的基原为菊科植物地胆草Elephantopus scaber L.。

Abstract:DMA fingerprinting among Elephantopus scaber L. ,E mollis . Pseudo - elephantopus spicatus and air commercial samples of Kudidan were dennonstrated with rantlomprimed polymerase chain reaction( PCR) including arbilranly - prined PCR(AP- PCR)and random were generated with two long (20,24mer)and one short (10mer)ra-nom - chosen primers. The three species ran be distinguished according to the banding patterns of their amplified DNA on agarose gels.The results showed that AP - PCR and RAPD fingerprints of the commercial Kudidan retailed in Fu-jian,Taiwan, Hong Kong and Macau markets matched that ot Elephantopus scaber I based on the Similarity indexes of the genonm DNA fingerprints.
