双黄益骨方治疗糖尿病骨代谢疾病的代谢组学研究

摘    要:目的:确定双黄益骨方治疗糖尿病骨代谢异常的作用位点,寻找相关的生物标志物,揭示中药治疗糖尿病骨代谢异常的治疗机制。方法:通过中药治疗糖尿病骨代谢异常患者的自身前后对照研究。运用核磁共振(1H-NMR)的代谢组学方法、系统生物学理论法,采用1H-NMR的代谢组学技术结合模式识别方法和SIMCA-P12.0统计分析治疗前后的代谢产物差异。结果:治疗前后的糖尿病骨代谢异常患者代谢产物存在显著差异:治疗前后相比,血液中的瓜氨酸、缬氨酸、甜菜碱、胆碱、脯氨酸、谷氨酰胺、O-乙酰糖蛋白、N-乙酰糖蛋白、丙酮酸、1-甲基组氨酸、三羧酸循环产物(α-酮戊二酸,柠檬酸)的水平上升,而支链氨基酸(亮氨酸、异亮氨酸)、葡萄糖、酪氨酸、丙氨酸的水平则下降。结论:通过上述代谢物的变化,提示双黄益骨方治疗糖尿病合并骨质代谢异常患者是通过改善机体代谢途径的紊乱,改善骨合成的原料及调控物质的调控来发挥功效。

Metabonomic Study on the Therapeutic Mechanism of Diabetic Osteoporosis with Shuanghuangyigu Recipe

Abstract:Objective: To determine the action site of the Chinese medicine of enriching kidney and promoting blood flow, find out the related biomarkers, and reveal the therapeutic mechanism of TCM. Methods: Through the self before-after study of Traditional Chinese Medicine in the treatment of patients with abnormal bone metabolism in diabetes, the metabolite differences before and after the treatment was analyzed with pattern recognition method and SIMCA-P 12.0, based on the method of nuclear magnetic resonance (1H-NMR) and biology theory. Results: There was a significant difference of metabolites in diabetic patients with abnormal bone metabolism before and after the treatment. Compared with the untreated, the levels of citrulline, valine, glycine betaine, choline, praline,glutamine, O-acetyl glycoprotens,N-acetyl glycoprotenis, pyruvic acid, 1-methyl histidine, three carbonaxylic acid cycle products'(α-ketoglutaric acid,citric acid) in blood all increased after the treatment, and the content of branched chain amino acids(leucine,isoleucine),tyrosine,alanine,glucose' in blood decreased. Conclusion: The Chinese medicine of enriching kidney and promoting blood flow can improve the metabolic disorders of the body, and regulate the content of bone synthesis of raw materials and chemicals.
