乙酰葛根素对大鼠脑缺血再灌注VEGF表达的影响

摘    要:目的探讨脑缺血再灌注后乙酰葛根素的脑保护作用及其机制。方法 线栓法制作脑缺血再灌注模型,分别对假手术组、脑缺血再灌注组、葛根素组、乙酰葛根素低(10mg·kg-1)、中(50mg·kg-1)、高剂量(250mg·kg-1)干预组实验标本作HE染色,采用免疫组化方法观察测定各组大鼠脑内的VEGF的表达情况并进行神经功能缺损评分。结果 脑缺血再灌注组与假手术组比较,VEGF表达增多(P〈0.01),药物干预组较缺血再灌注组VEGF表达均增多(P〈0.01),且神经元损伤较轻,且三个剂量的表达有显著差别(P〈0.01)。结论 乙酰葛根素对脑缺血再灌注有明显的保护作用,诱导VEGF表达可能是其对脑缺血再灌注损伤起保护作用的机制之一。

Abstract:Objective To investigate the protective effect of acetylpuerarin on cerebral ischemia-reperfusion injury in rats and itsmechanism. Methods Wistar rat was used as a model of focal cerebral ischemia and reperfusion to observe expression of VEGF after focal cerebral ischemia-reperfusion and the protective function of three levels of doses Acetylpuerarin. Immunohistochemicalmethod were used to measure the number of VEGF positive cells in rat’s cerebral cortex of sham-operated group,model group and Acetylpuerarin group. Results VEGF had a high-level expression and increased significantly after ischemia reperfusion(P<0.01).The Acetylpuerarin can significantly increase the expression of the VEGF,and the highest expression after the highest Acetylpuerarin group (P<0.01).Conclusion The Acetylpuerarin had obviously protective function for ischemia-reperfusion rat.
