益气养阴方对Graves病合并白细胞减少小鼠IL-13表达影响的实验研究

目的观察Graves病（GD）小鼠外周血白细胞计数和小鼠细胞因子IL-13表达及益气养阴方对其的影响。方法基因枪造模建立GD小鼠模型,应用中药益气养阴方治疗后,观察小鼠外周血白细胞计数和小鼠细胞因子IL-13表达的变化。结果治疗30天后,在外周血白细胞计数方面,中药益气养阴方高剂量组明显高于中药益气养阴方低剂量组、中剂量组（P〈0.01）;在降低甲状腺激素水平方面,中药益气养阴方高、中剂量组明显优于中药益气养阴方低剂量组;在小鼠细胞因子IL-13表达方面,西药他巴唑组小鼠细胞因子IL-13表达显升高,与正常对照组和模型组比较有显著差异性（P〈0.01）。中药治疗组的IL-13mRNA表达与正对照组比较没有显著性差异,P〉0.05。经中药益气养阴方治疗,小鼠IL-13mRNA的表达下调,即中药高剂量组表达最低,依次增高。结论益气养阴方对Graves病合并白细胞减少有较好的治疗作用,其机理可能在于其能较好地调节Graves病小鼠细胞因子IL-13水平,有促进免疫细胞成熟、活化、增殖和免疫调节的作用。

Experimental Study in Effect of Yiqiyangyin Decoction on IL-13of GD Combining With Leukopenia Mice

Objective To observe the effect of Yiqiyangyin decoction on the peripheral white blood cell count and IL-13 expression of GD Mice To discuss the mechanism of yiqiyangyin decoction to merger leukopenia. Methods To establish the model of GD mice by Gene gun.After the therapy of Yiqiyangyin decoction,we observed the peripheral white blood cell count and IL-13 expression alteration of GD Mice.Results After the treatment in 30 days,the peripheral white blood cell count in high-dose group of Yiqiyangyin decoction was significantly higher than low-dose group and middle-dose group（P〈 0. 01）. High-dose group and middle-dose group of Yiqiyangyin decoction was significantly superior to low-dose group in reducing thyroid hormones,. The tapazole group decreased obviously,which was significant difference with normal group in IL-13 expression（P〈 0. 01）. There was no significant difference in IL-13 mRNA expression between chinese medicine treatment group and normal group（P〉 0. 05）. After the treatment of Yiqiyangyin decoction,the IL-13 mRNA expression down regulated,which mean High-dose group express the lowest,with gradually increasing. Conclusions Yiqiyangyin decoction has a better therapeutic effect to Graves disease combining with leukopenia. Its mechanism may be related with better regulating IL-13 expression.Yiqiyangyin decoction can promote the maturation,activation,proliferation,and immunodulatory of immune cell.
