复方黄柏液对内皮细胞增殖迁移成管影响的机制研究

目的:探索复方黄柏液对内皮细胞增殖迁移成管影响的机制,为复方黄柏溶液用于治疗糖尿病足溃疡提供依据。方法:用CCK-8法检测药物毒性，探索复方黄柏液使用的最佳药物浓度、CCK-8法检测细胞增殖活力、细胞划痕实验检测细胞迁移率、成管实验检测细胞成管能力、蛋白质印迹法(Western Blotting)实验检测蛋白表达水平的变化。结果:使用复方黄柏液后,高糖组内皮细胞的增殖、迁移、成管能力较前有了提高,并降低了促血管生成素2(Ang-2)蛋白水平,增加血管内皮生长因子(VEGF)和磷酸化酪氨酸激酶受体-2(P-Tie)蛋白水平。结论:复方黄柏液通过促进内皮细胞增殖、迁移、成管以及影响蛋白表达水平,改善内皮功能障碍来实现对糖尿病足溃疡的恢复。

Effect and Mechanism of Fufang Huangbai Liquid on Proliferation,Migration,and Tube Formation of Endothelial Cells

To explore the effect and mechanism of Fufang Huangbai Liquid on endothelial cell proliferation,migration,and tube formation and provide evidence for the treatment of diabetic foot ulcer by Fufang Huangbai Liquid.Methods:Cell Counting Kit-8(CCK-8) was used for the detection of drug toxicity to determine the optimal concentration of Fufang Huangbai Liquid.Cell proliferation activity was detected by CCK-8 assay,cell migration rate by wound healing assay,tube formation ability by tube formation assay,and protein expression changes by Western blot.Results:After Fufang Huangbai Liquid treatment,the proliferation,migration,and tube formation abilities of endothelial cells in the high-glucose group were potentiated.Furthermore,the angiopoietin 2(Ang2) protein expression was reduced,and the protein expression levels of vascular endothelial growth factor(VEGF) and phosphorylated tyrosine kinase receptor-2(p-Tie2) were up-regulated.Conclusion:Fufang Huangbai Liquid can relieve diabetic foot ulcer by promoting endothelial cell proliferation,migration,and tube formation,affecting protein expression,and improving endothelial dysfunction.
