涤痰开窍汤对脑卒中后抑郁大鼠行为能力及脑内神经递质的影响

摘    要:目的 探讨涤痰开窍汤对脑卒中后抑郁(PSD)模型大鼠行为能力及脑内神经递质的影响.方法 采用双侧颈总动脉永久性结扎后予以行为限制制备PSD大鼠模型,随机分为模型组、涤痰开窍组、氟西汀组,并设假手术组,采用RP-HPLC-荧光检测法测定大鼠海马区单胺类神经递质的变化,并观察大鼠自发性行为改变.结果 模型组大鼠行为能力下降(P<0.01或P<0.05),海马区5-羟色胺、5-羟基吲哚乙酸、多巴胺、去甲肾上腺素水平下调(P<0.05).经盐酸氟西汀、涤痰开窍汤治疗后大鼠的水平活动、垂直活动及清洁活动均较模型组改善,脑内单胺类神经递质水平提高(P<0.05).结论 大鼠双侧颈总动脉永久性结扎结合行为限制可为卒中后抑郁的实验及临床研究提供较为理想的动物模型.涤痰开窍汤可以改善脑卒中后抑郁大鼠的行为能力及脑内单胺类神经递质水平.

Abstract:Objective To study the effect of Ditan Kaiqiao Decoction on the behavioral ability and intracerebral neurotransmitter of rats with post-stroke depression(PSD). Methods After the permanent ligation of bilateral common carotid arteries was performed, the behavioral limitation was applied to make PSD model rat and they were randomized into Model group, Ditan Kaiqiao group, Fluoxetine group and Pseudooperation group. The changes of monoamine neurotransmitter of hippocampal region were tested by RP-HPLC fluoroscopic examination and the autonomy behavioral changes were observed. Results The behavioral ability of Model group decreased (P<0.01 or P<0.05). 5-HT, 5-HIAA, DA and NE of hippocampal region were lowered (P<0.05). With the treatment of fluoxetine hydrochloride and Ditan Kaiqiao Decoction, the horizontal movement, vertical movement and clean movement of rats were improved in comparison with Model group, while the intracerebral monoamine neurotransmitter increased (P<0.05). Conclusion The permanent ligation of bilateral common carotid artery in combination of behavioral limitation could provide the ideal animal models for experiment and clinical study of PSD. Ditan Kaiqiao Decoction could improve the behavioral ability and intracerebral neurotransmitter of PSD rats.
