防哮饮早期干预对哮喘小鼠嗜酸性粒细胞和气道炎症的影响

摘    要:目的 观察防哮饮早期干预对哮喘气道炎症的防治作用机制. 方法 将50只BALB/c健康小鼠随机分为正常对照组、哮喘模型组、防哮饮小剂量组、防哮饮大剂量组、布地奈德(BUD)组,每组10只,用卵蛋白(OVA)致敏激发造模.防哮饮小剂量组、大剂量组从小鼠第1次致敏当天起分别灌胃给予防哮饮500g/L、2 500g/L,每天1次,每次0.4ml,共26天.正常对照组和哮喘模型组则以等量生理盐水灌胃.BUD组于第23天予BUD溶液(0.5g/L)0.01ml/d,滴鼻3天.最后1次激发后48h处死小鼠,检测小鼠血清白细胞介素4(IL-4)、白细胞介素5(IL-5)、γ-干扰素(IFN-γ)、白细胞介素12(IL-12)的含量,光镜下观察肺部EOS浸润和骨髓EOS百分计数(EOS%).结果 哮喘模型组小鼠肺组织HE染色显示小鼠肺部出现大量的炎症细胞浸润,以EOS和中性粒细胞为主.哮喘模型组骨髓EOS%、血清IL-4/IFN-γ的比值、IL-5/IL-12的比值较正常对照组明显升高(P<0.01);防哮饮大剂量组和BUD组均能显著降低骨髓EOS%、明显抑制气道及血管周围炎症浸润、降低哮喘小鼠血清IL-4/IFN-γ和IL-5/IL-12的比值(P<0.05或P<0.01),两组比较差异无统计学意义(P>0.05). 结论 防哮饮能有效下调哮喘小鼠血清IL-4/IFN-γ和IL-5/IL-12的比值,降低骨髓EOS%,抑制以EOS浸润为主的气道炎症.

Abstract:Objective To investigate the effect and mechanism of early intervention with Fangxiaoyin Granules (Granules for preventing asthma) on airway inflammation in asthmatic mice.Methods Fifty healthy BALB/c mice were randomized into 5 groups,i.e.normal control group,asthma model group,Fangxiaoyin Granules small-dose group,Fangxiaoyin Granules large-dose group,and budesonide (BUD) group,with 10in each.The asthmatic models were established with ovalbumin (OVA) sensitization for inspiration.The Fangxiaoyin Granules small-dose group and large-dose group were given small-dose (500g/L crude medicine) and large-dose (2500g/L crude medicine) of Fangxiaoyin Granules respectively through gastric feeding from the first day of sensitization,once a day,0.4ml each time,26days in all.The normal control group and asthma model group were given normal saline in the equal amount through gastric feeding.The BUD group was given BUD solution (0.5g/L) for nasal dripping since the 23rd day under the ethyl ether inhalation anesthesia,0.01ml/day,for 3days.Forty-eight hours after the last inspiration,the mice were put to execution.The contents of serum interleukin-4 (IL-4),interleukin-5 (IL-5),interfer-on-γ (IFN-γ),and interleukin-12 (IL-12) were measured,and the eosinophils (EOS) infiltration in the lung and marrow EOS% were observed with photoelectric microscope.Results In the asthma model group,the HE dyeing of the lung tissues showed that there were massive inflammatory infiltration,mainly EOS and neutrophils;the marrow EOS%,serum IL-4/IFN-γratio,and IL-5/IL-12ratio were significantly increased in comparing with those of the normal control group (P<0.01).In the Fangxiaoyin Granules large-dose group and BUD group,the marrow EOS% was significantly decreased,the airway and perivascular inflammatory infiltration was significantly inhibited,and serum IL-4/IFN-γratio and IL-5/IL-12ratio were significantly decreased,the difference between the two groups was not statistically significant (P>0.05).Conclusion Fangxiaoyin Granules could effectively down-regulate serum IL-4/IFN-γratio and IL-5/IL-12ratio,decreasing the marrow EOS%,and inhibiting the airway inflammation mainly with EOS infiltration.
