茵陈五苓散抗大鼠动脉粥样硬化作用机理探讨

摘    要:目的探讨茵陈五苓散对动脉粥样硬化大鼠的作用机理。方法70只雄性Wistar大鼠随机分为正常组、模型组、茵陈五苓散组、绞股蓝组。采用高脂饲料喂饲法复制大鼠动脉粥样硬化模型,治疗1个月后分别检测血脂、血液流变学、主动脉组织学及超微结构变化、血管平滑肌细胞(VSMC)bcl-2 mRNA的表达。结果茵陈五苓散降低总胆固醇、甘油三酯、低密度脂蛋白胆固醇,升高高密度脂蛋白胆固醇,降低血液黏度、红细胞压积、血小板黏附率,维持主动脉组织结构,下调基因bcl-2 mRNA的表达均优于绞股蓝。结论茵陈五苓散具有良好的抗动脉粥样硬化作用,其下调相关基因bcl-2 mRNA的表达可能是其作用机理。

Abstract:Objective To study the effect and mechanism of Yinchen Wuling Powder(Powder of Oriental Wormwood and Five Herbs with Poria) on atherosclerosis(AS) rats.Methods Totally 70 Wistar rats were randomized into normal group, model group, Yinchen Wuling Powder group, and Gypenosides group.The AS models were made by feeding greasy food for producing AS plague in aorta.The serum lipid,blood rheology,uhrastructure of aorta tissues,and bcl-2 mRNA expression of VSMC were tested after one month to see the changes.Results Yinchen Wuling Powder could obviously decrease the total cholesterol,triglyceride,and low den- sity lipoprotein cholesterol;increase the high density lipoprotein cholesterol;reduce the blood viscosity,packed cell volume,and platelet adhesion rate;maintain the aorta structure;and lower the bcl-2mRNA expression.The effect was better than that of Gypeno- sides.Conclusion Yinchen Wuling Powder has a significant effect on AS and its functional mechanism may lie in lowering the bcl-2 mRNA expression.
