| 摘 要： | 目的]探讨脾虚大鼠结肠上皮细胞水通道蛋白8（AQP8）表达的变化规律，为脾虚泄泻的发生机制提供实验依据。方法]以RT-PCR及免疫组化等方法检测脾虚模型大鼠结肠AQP8mRNA表达及蛋白含量，光镜观察黏膜组织改变。结果]脾虚模型组大鼠出现腹泻、少食、消瘦等表现；结肠黏膜上皮不完整，细胞排列疏松；AQP8 mRNA表达及蛋白含量均比正常对照组减少（P〈0．05，〈0．01）。结论]AQP8表达下调可能是脾虚大鼠产生泄泻病理生理机制之一。

| Abstract: | Objective]Objective： to explore the aquaporin 8 （AQP8） expression in colonic epithelium of rats with spleen-deficiency and to provide experimental evidence for the mechanism of spleen deficiency. Methods] Immunohistochemistry and RT-PCR were employed to detect the AQP8 protein and mRNA expression in colonic epithelium, and colonic mucosa morphology was observed with light microscope. Results] Compared with rats in the control group, the rats in spleen deficiency group had diarrhea, less eating and loss of body weight, and meanwhile, colonic mucosa was not integrated, rarely arrayed; AQP8 protein and its mRNA expressions decreased markedly （P〈0. 05,〈0.01）. Conclusion ]Downregulation of AQP8 expression might be one of the pathophysiologic mechanisms of diarrhea in spleen-deficiency rats. |
