摘    要:目的探讨d-δ-三烯生育酚抗鼻咽癌的生物学活性。方法将低分化5-8F鼻咽癌细胞随机分为观察组和对照组,观察组分别予40、50、60panol/Ld-δ-三烯生育酚干预48h,对照组不予干预。采用MTT实验、流式细胞仪检测细胞增殖抑制率、凋亡率及细胞周期;荧光染色和显微镜观察细胞形态学变化。结果观察组干预48h后,随浓度增加,镜下观察活细胞减少,漂浮的死亡细胞增多,细胞核强荧光染色细胞比例增多,细胞出现明显的抑制和凋亡现象。观察组不同浓度细胞抑制率及凋亡率明显升高,且明显高于对照组(P均〈0.01);对照组S期的细胞百分率明显低于观察组,P均〈0.01。结论d-δ-三烯生育酚能抑制5--8F细胞增殖,诱导其凋亡,具有潜在的抗鼻咽癌生物学活性。

Abstract:Objective To investigate the biological effects of d-δ-tocotrienols against nasopharyngeal carcinoma. Methods Nasopharyngeal carcinoma cell line 5-8F was cultured and divided into control group and observed group , the control group was not intervened,the observed group were treated with d-δ-toeotrienols of 40,50,60 mol/L respectively for 48 hs. The inhibition rate of cell proliferation, apoptosis rate and cell cycle were detected with MTT experiment and flow eytometry;the changes of cell morphology were observed ,with fluorescent staining and microscope. Results After the treatment for 48 hs, the living cells reduced, the float dead cells increased, the cell proportion with nucleus strong fluorescence dyeing increased, the cells presented obvious suppression and apoptosis with the augment of d-δ-toeotricnols. Compared with the control group, cell suppression and apoptosis rates increased significantly in the observed group,both P 〈 0.01 ; S- phase cell percentage of control group was significantly lower than that of the observed group (P 〈0.01 ). Conclusions d-δ-tocopherol has powerful ability to inhibit proliferation of 5-8F cell line and induce its apoptosis, and posses potential biological effects against nasopharyngeal carcinoma.
