摘    要:目的: 探讨聚乳酸-聚乙醇酸(PLGA)与骨髓间充质干细胞(BM-MSCs)衍生的心肌样细胞的生物相容性,为构建心肌组织工程及其进一步体内回植提供研究基础。方法: 以密度梯度离心法体外分离培养SD大鼠的BM-MSCs,将培养的第3 代细胞用终浓度为10 μmol/L的5-氮杂胞苷和0.1μmol/L的血管紧张素Ⅱ诱导分化。将诱导后的心肌样细胞接种到PLGA材料上, 用沉淀法测定细胞的黏附力和黏附率;MTT比色法检测细胞的增殖并绘制增殖曲线;扫描电镜观察细胞在PLGA材料上的形态学特征及细胞与该材料的相容性。结果: 心肌样细胞在PLGA支架材料上贴附、生长良好,可分泌细胞外基质,其黏附、增殖能力与单纯的细胞相比无显著统计学差异。结论: PLGA与心肌样细胞具有良好的生物相容性,可作为心肌样细胞的理想载体构建心肌组织工程。

Abstract:AIM: To investigate the biocompatibility of polylactic-co-glycolic acid (PLGA) and cardiomyocyte-like cells derived from bone marrow mesenchymal stem cells (BMMSCs) so as to provide a basis for the study of myocardial tissue engineering and in vivo implantation. METHODS: BMMSCs were isolated from bone marrow of SD mice by density gradient centrifugation. BMMSCs of passage 3 were induced and differentiated to cardiomyocyte-like cells with 10 μmol/L 5-azacytidine (5-aza) and 0.1 μmol/L angiotensin II (AngII) in culture medium. Cardiomyocyte-like cells were then inoculated into the pores of PLGA pieces. The cell adhesion rate and the adhesive power were examined by conventional precipitation method and the proliferation behavior of the cells was analyzed by MTT assay. Scanning electron microscopy (SEM) was used to observe the morphology of cardiomyocyte-like cells. RESULTS: Cardiomyocyte-like cells attached, proliferated and secreted extracellular matrix in porous PLGA. No obvious significance was observed between PLGA group and cell-only group. CONCLUSIONS: Cardiomyocyte-like cells can adhere to PLGA and proliferate, indicating PLGA can be used as a scaffold in myocardial tissue engineering.
