摘    要:采用钌红染色电镜观察法对正常和高脂饲食兔主动脉内膜蛋白多糖(PG)的变化进行了比较研究。结果表明正常和病变的内膜能看到两型钌红颗粒。但病变含脂区钌红颗粒数量明显减少。正常内皮细胞表面有一层钌红阳性外衣,高脂饲食20天后这层外衣开始变薄,90天几乎消失;平滑肌细胞(SMC)膜表面亦有一层钌红阳性外衣,当SMC变成泡沫细胞后,这层外衣也变薄或消失。本文讨论了内膜PG的变化与动脉粥样硬化(AS)发病学的关系。认为动脉壁PG代谢紊乱是AS发病的重要原因之一。

Abstract:The distribution and ultrastrctural changes of proteoglycan in aortic intimal of dietary in- duced atherosclerosis in rabbits were studied qulitatively with electron microscope. Two types of ruthenium red-positive granules were demonstrated in the intercellular matrix both in normal and atherosclerotic inti- mals, but in the lipid containing areas, these qranules were found markedly diminished. The ruthenium red positive coat Which was present on the luminal surface of normal endothelial cells began to appear thinner af- ter 20 days of high fat-feeding and became almost disappeared in the 90 days. Ruthenium red staining ma- terial could be observed as a thin coat lying along the outer surface of the smooth muscle cell (SMC), but after the SMC transformed into foam cell (FC), it became thin or disappeared. The correlations between proteo- glycan and endothelial permeablity and that between proteoglycan and pathogenesis of AS were discussed. The results of this study suggest that metabolic disorder of proteoglycan is one of the important etiologic fac- tors of atherosclerotic pathogenesis.
