槲芪癥消汤对人肝癌细胞株HepG2.2.15增殖和凋亡的影响

目的:研究槲芪癥消汤对体外培养的人肝癌细胞株HepG2.2.15的增殖、凋亡、迁移能力、细胞内p53基因及蛋白表达量的影响,揭示该方可能的抗肿瘤机制。方法:应用不同浓度的槲芪癥消汤处理细胞,MTT法测定细胞增殖抑制率,Transwell检测细胞侵袭能力、流式细胞术检测细胞凋亡情况、rt-qPCR及Western blot检测细胞内p53的RNA及蛋白的表达变化。结果:槲芪癥消汤对HepG2.2.15细胞增殖有明显的抑制作用,使细胞的侵袭能力明显下降,并可促进细胞凋亡,同时细胞内p53的RNA及蛋白表达量明显增加,与对照组比较,差异均有统计学意义(均P<0.05)。结论:槲芪癥消汤能抑制肝癌HepG2.2.15细胞的增殖、侵袭,促进其凋亡,可能与促进细胞内的p53基因表达有关。

The effect of Hiqizhengxiao decoction on proliferation and apoptosis of human hepatocellular carcinoma cell line HepG2.2.15

Objective:Through the study of the impact of Huqizhengxiao decoction and its effective components on HepG2.2.15cell belong to hepatocellular carcinoma cell line,about its proliferation,apoptosis and migration,the expression of p53 gene and protein in cells,to reveal the possible mechanism of its tumor-inhibitive effect.Methods:Use different concentrations of“huqizhengxiao”decoction to deal with the HepG2.2.15 cells cultured in vitro,then observe the proliferation of HepG2.2.15 cells by MTT method,use Transwell chamber to detect cell migration,Flow cytometry to detect Cell apoptosis、Fluorescence quantitative and Western blot to detect the expression of p53 gene and protein in cells.Results:Huqizhengxiao decoction can obviously inhibit the proliferation of HepG2.2.15cells,decrease the invasive ability,promote the apoptosis.At the same time,the expression of p53 RNA and protein in HepG2.2.15 cells is significantly increased,compared with the control group,the difference is significant(P<0.05).Conclusion:“Huqizhengxiao”decoction can inhibit the proliferation and migration of HepG2.2.15 cells,increase the cell apoptosis.The expression of p53 gene and protein increases more obviously in the“Huqizhengxiao”decoction group,which indicates that p53 pathway may be one of the anticancer mechanism of"Huqizhengxiao"decoction.
