摘    要:目的 为获取中华按蚊感染约氏疟原虫后24 h的高表达基因热激蛋白40(HSP40)cDNA的全长序列. 方法 运用抑制性消减杂交技术(suppression substractive hybridization,SSH)构建中华按蚊感染约氏疟原虫后24 h的差异表达基因文库,从高表达文库中获得长度为363 bp的HSP40基因的cDNA片段,运用RACE技术对其cDNA序列进行全长扩增. 结果 获得中华按蚊HSP40 cDNA的全长序列,总长度为1 159 bp,开放阅读框为1 014 bp,编码337个氨基酸. 结论 获得了中华按蚊HSP40 cDNA的全长序列.

Abstract:Objective To isolate differentially expressed gene HSP40 from Anopheles sinensis induced by infection with Plasmodium yoelii yoelii after 24 h, and to amplify the full-length cDNA of HSP40. Methods A 363 bp of HSP40 cDNA was isolated from the suppression substractive hybridization (SSH) library. The full length of cDNA was amplified using the RACE (rapid-amplification of cDNA ends) technology. Results The full-length cDNA of HSP40 was completely amplified. The whole length was 1 159 bp, with an open reading frame of 1 014 bp, encoding 337 amino acids. Conclusions The full-length cDNA of HSP40 has been completely amplified.
