广西凭祥市和宾阳县HIV-1流行毒株gag基因的序列测定和亚型分析

目的了解广西壮族自治区凭祥市和宾阳县吸毒人群中流行的艾滋病病毒1型(HIV-1)毒株的亚型及各种亚型的分布特点。方法对HIV-1阳性的10例吸毒人员和1例受血者进行流行病学调查,采集外周静脉抗凝血,提取前病毒DNA进行体外扩增,并对获得的gag区基因的核酸片段进行测序和分析。结果宾阳县的6份样品为CRF08-BC重组株,凭祥市的4份样品均为CRF01-AF重组株,一份输血引起的样品为B’亚型。结论在凭祥市和宾阳县吸毒人群中流行的是不同的HIV-1重组株,而且在同一地区的亚型之间进化比较接近。以上资料表明,CRF08-BC和CRF01-AE在这两地区的静脉吸毒者中成为主要的流行株。

Sequencing of Gag gene of prevalent HIV-1 strains and their subtypes in some districts of Guangxi

Objective To study the distribution of subtypes of human immunodeficiency virus-1(HIV-1) strains prevalent in Pingxiang City and Binyang County of the Guangxi Zhuang Autonomous Region in order to predict their epidemic trends.Methods Epidemiological investigation was carried out among ten HIV-1 positive drug users and one HIV-1 positive former blood donor and their DNA fragments of HIV-1 gag region were amplified by nested-PCR.All these fragments were sequenced.Results Sequencing analysis showed that strains isolated from IDUs in Pingxiang and Bingyang were all HIV-1 circulating recombinant forms(CRFs),i.e.CRF08-BC(n=6) and CRF01-AE(n=4),respectively;and the strain isolated from the former blood donor was HIV-1 strain B' subtype.Conclusion There were two different CRFs of HIV-1 among IDUs and one B' strain of HIV-1 among the former blood donor.The phylogenetic tree analysis showed that the strains isolated from Pingxiang and Bingyang were indistinctive variation,and that CRF08-BC and CRF01-AE might represent the main strains circulating in Guangxi.
