急、慢性病毒性心肌炎小鼠辅助T淋巴细胞亚群动态观察及药物干预的影响

摘    要:目的 :观察 BAL B/ c小鼠在感染柯萨奇病毒后辅助 T淋巴 (Th)细胞—— Th1 、Th2 细胞的动态变化以及抗柯注射液 (RSF)的干预治疗对其影响。方法 :将 15 6只 BAL B/ c小鼠分别制备成急性心肌炎、慢性心肌炎模型 ,各模型随机分为正常组、病毒对照组和 RSF治疗组 ,用 EL ISA法检测各组小鼠 Th1 、Th2 类细胞因子。结果 :在急性心肌炎模型中 ,病毒对照组在感染后第 3天 Th1 达到高峰 ,后逐渐下降 ,到第 12天仍高于正常 ,而 Th2 基本于正常范围 ;RSF治疗组除第 3天外 ,Th1 均下降 ,Th2 则表现为有明显的触发效应。慢性心肌炎模型中 ,各组Th1 与 Th2 类细胞因子均增高。结论 :在急、慢性心肌炎中 ,Th1 类细胞在病程中起着主导地位 ,Th2 类细胞则有利于疾病的改善。 RSFA对 cox B3m心肌炎有显著的治疗作用 ,其作用机制可能是通过对 Th亚群的调节来实现——即抑制 Th1 细胞 ,增高 Th2 细胞的表达来进行免疫调节。

Abstract:Objective:Study about the regulatory mechanism of the characteristic cytokine products of Th 1 and Th 2 cells and the expressions of Th 1,Th 2 cell intervened by Radix sophora flavescens (RSF).Method:To prepare the acute and chronic myocarditis of the murine model (156 cases).They were divided into 3 groups at random,such as CVB 3m infectious group,RSF therapeutic group and control group.The characteristic cytokines (IL 2 and IL 10) of the subsets were detected by ELISA kits (pharmingen co).Result:Th 1 like cytokine IL 2 was mostly expressed in the acute myocarditis,it was elevated maximal on day 3,and subsequently decreased,still higher than normal on day 12.The Th 2 like cytokine IL 10 was in normal range all along.In the chronic myocarditis model,the expression of cytokines was enhanced in both Th 1 and Th 2 like cells.Conclusion:The immunophysiologic responses in acute and chronic myocarditis be mainly induced by Th 1 cells,and Th 2 cells help inhibit inflammation,RSF has therapeutic roles in CoxB 3m myocarditis by regulating T help cells subsets level it facilated in inhibiting Th 1 cells and enhancing the expressions of Th 2 cells.
