维生素C对光照后兔视网膜色素上皮细胞内B细胞淋巴瘤表达的影响

摘    要:目的观察维生素C(Vit C)对光照后兔视网膜色素上皮(retinal pigment epithelium,RPE)细胞内B细胞淋巴瘤(Bcl-2)表达的影响。方法使用酶消化法体外分离培养色素兔的RPE细胞,用可见光照射造成其凋亡。观察不同剂量的维生素C(Vit C)在不同的给药时间对兔RPE细胞Bcl-2表达的影响。结果色素兔RPE细胞经可见光照射后细胞内Bcl-2均有不同程度的表达,其表达数量与Vit C剂量呈正相关。结论Vit C可以增加光照后兔RPE细胞Bcl-2的表达,它可能起到抑制光损伤、减少凋亡的作用。

Abstract:Objective To observe the expression of Bcl-2 when retinal pigment epithelial cells of the pigmented rabbit are exposed to visible light and to observe the effect that vitamin C has on this expression.Methods Retinal pigment epithelial cells of the pigmented rabbit were collected from enzyme digested retinas and cultured in vitro. Apoptosis of the cultured retinal pigment epithelial cells was caused by exposing the cells to visible light. The expression of Bcl-2 in the cultured cells was examined in the presence of different doses of vitamin C that had been administered at different time points.Results Bcl-2 was expressed for different lengths of time after the cultured RPE cells of the pigmented rabbits had been exposed to visible light. Expression is positively correlated to the amount of vitamin C introduced in the cell culture.Conclusion The application of vitamin C can increase the expression of Bcl-2 when retinal pigment epithelia cells are exposed to visible light. We speculate that vitamin C can inhibit photic injury of the retina and prevent cell apoptosis.
