大麻素对牛眼小梁细胞增殖、粘附和迁移的影响

摘    要:目的:通过研究内源性大麻素物质(anandamine,AEA),对体外培养的牛眼小梁细胞的增殖、黏附以及迁移功能的影响,探讨AEA降低眼压的机制。方法:对牛眼小梁细胞进行体外原代培养及传代培养,应用免疫组化的方法(NES,Ⅷ因子相关抗原染色)鉴定细胞,化学及透射电镜观察。对3代牛眼小梁细胞分别以含AEA10,1,0.1,0μmol/L DMEM(含100ml/L小牛血清)培养液进行干预,分别于24,2h,2h后用MTT法计数细胞,再利用计算机统计软件进行数据分析,观察其对牛眼小梁细胞的增殖、粘附及迁移功能的影响。结果:内源性大麻素,在一定浓度范围内,对体外培养的牛眼小梁细胞的增殖、粘附及迁移功能均有较为明显的抑制作用(P<0.05)。结论:由此,可以了解到AEA可能是通过影响小梁细胞的增殖、粘附及迁移功能来改变房水流出途径的阻力,而降低眼内压。

Abstract:AIM: To study the effect of endogenous cannabinoid (Anandamine, AEA) on proliferation, adherence and immigration in cultured bovine trabecular cells (BTCs) and to investigate the mechanism of AEA on the degrading intraocular tension.·METHODS: BTCs were primarily cultured and subcul-tured, detected by NES and observed under TEM. The third passage cells were incubated with different dosages of AEA (10μmol/L, 1μmol/L, 0.1μmol/L, 0μmol/L, final concentration, diluted by DMEM) for 24, 2 and 2 hours. The number of BTCs was counted with MTT method. Then the results were analyzed by the computer data-analysis system and statistical test.·RESULTS: AEA inhibits the proliferation, adherence and immigration in BTCs, in comparison and control group(P<0.05).·CONCLUSION: On this data, AEA possibly degrades intraocular tension through affecting the outflow in trabecular meshwork.
