视网膜感光细胞纯化方法的研究

摘    要:目的 探讨改进获取视网膜纯感光细胞的方法。方法 应用改良的准分子激光切削法和酶分层消化法来分离纯化视网膜感光细胞层。结果 由两种方法获取的视网膜组织片经苏木素 -伊红染色均证实为单一的纯感光细胞层并保持生物活性。结论 改良的准分子激光切削法和酶分层消化法均能获取视网膜纯感光细胞 ,且对细胞损伤小 ,内外节段保持完好 ,可为视网膜的移植创造条件。

Abstract:ObjectiveTo investigate methods for isolation of the pure retinal photoreceptors.MethodsModified excimer laser surgery,regular excimer laser surgery, and enzyme gradation digestion were used to isolate retinal photoreceptors.The resulting retinal tissues were stained with hematoxylin eosin(HE).ResultsThe retinal sheets obtained by these three methods each demonstrated a layer of pure photoreceptors by HE staining.Modified and regular excimer laser surgery could both precisely ablate retina.Modified excimer laser ablation obtained photoreceptor sheets with perfect inner and outer segment,while regular excimer laser ablation got photoreceptor sheets with impaired inner and outer segment.Enzyme gradation digestion could also result in photoreceptor sheets with good inner and outer segments,although there was non precise digestion.ConclusionModified excimer laser surgery and enzyme gradation digestion can be used to obtain a layer of pure photoreceptors with good tissue preservation;specially perfect inner and outer segment.These cells may be a source for retinal transplantation.
