基质金属蛋白酶抑制剂治疗角膜新生血管的实验研究

摘    要:目的:观察人工合成基质金属蛋白酶抑制剂点眼对角膜新生血管的影响。方法:15只新西兰白兔双眼角膜植入脂多糖-醋酸乙烯聚合膜造成角膜新生血管模型后,治疗眼点基质金属蛋白酶抑制剂GM6001,对照眼点HEPES缓冲液,连续用药14日,处死动物摘下角膜做光、电镜观察。结果:GM6001点眼显著地抑制角膜新生血管的生长并减轻角膜的炎症反应。组织病理学检查显示新生血管化的角膜实质内有大量炎性白细胞浸润。结论:提示局部应用人工合成基质金属蛋白酶抑制剂对角膜新生血管有一定疗效,但治疗机制有待进一步研究。

Abstract:ObjectiveTo evaluate the effect of topical application of GM6001,a synthetic metalloproteinase inhibitor on corneal neovascularization.MethodsCorneal neovascularization was induced in both eyes of 15rabbits by surgical implantation of an ethylene vinyl acetate copolymer disk (2mm in diameter) containing 10% lipopolysaccharide into the corneal lamellae.The treatment group received topical GM6001 six times a day, while the control received the vehicle in the same way. After a 2-week treatment,corneal neovascularization was evaluated in a double-masked manner according to a scoring system and then,the eyes were removed for pathological examination.ResultsGM6001 significantly prevented corneal neovascularization compared with the vehicle.Histopathologically,the eyes treated with GM6001 had a less inflammatory response than those with vehicle.ConclusionGM6001 may be a therapeutically useful angiostatic agent for corneal neovascularization.As yet, the precise mechanisms for its angiostatic activities remain to be elucidated.
