Abstract:Objective:To investigate the correlation between prostate epithelium-specific Ets transcrip-tion factor(PDEF) expression and Gleason grade and clinical stage in hormone-independent prostate can-cer(PCa).Methods:Thirty androgen-dependent prostate cancer specimens(group A) and 30 androgen-independent prostate cancer specimens(group B) were collected in this retrospective study.PDEF gene and protein expression was detected with immunohistochemical SP method,RT-PCR semi-quantitative method and Western blotting method.The correlation between PDEF expression and Gleason grade,clinical stage was analyzed.Results:Immunohistochemical results showed that PDEF positive expression rate was 70%(21/30) in group A,while that was 96.7%(29/30) in group B(P0.05).PDEF positive expression rate in cases with ≤7 points in Gleason grade was significantly lower than that in the cases with 7 points(64.3% vs 93.7%,P0.01).PDEF positive expression rate in the cases with T2 in clinical stage was significantly higher than that in the cases with ≤ T2(86.8% vs 54.5%,P0.01).RT-PCR experiments further confirmed that there was a significant difference in PDEF mRNA expression between group A and group B(P0.01).PDEF mRNA expression in the cases with ≤7 points in Gleason grade was significantly lower than that in the cases with 7 points(P0.01).PDEF mRNA expression in the cases with T2 was significantly higher than that in the cases with ≤ T2(P0.01).Western blotting result showed that PDEF protein expression was as the same as PDEF mRNA.Conclusion:PDEF was highly expressed in androgen-independent prostate cancer specimens,and further up-regulated with the increase of clinical stage and Gleason grade,indicating that PDEF might involve in the occurrence and development of PCa,and thus PDEF might be a target gene in the study and treatment of hormone-independent PCa.
