牛磺酸对小鼠抗氧化和DNA修复能力的作用

摘    要:断乳小鼠在饲料相同的基础上,两组动物分别喂以普通水和1%牛磺酸,于36天(幼龄期)和110天(中龄期)时断头处死,测定其肝、脑匀浆的SOD活性,MDA及脂褐质含量,结果表明:补充牛磺酸的小鼠肝脑匀浆SOD在幼龄期即显著高于对照组(P<0.05),至中龄期更为显著(P<0.01)。脑匀浆中MDA及脂褐质含量在幼龄期时无差异,至中龄时,实验组含量显著低于对照组(P<0.01)。肝匀浆中脂褐质含量在不同鼠龄时与脑匀浆相同,但MDA则不同鼠龄时无变化。本实验又以110天、150天、180天鼠龄之脾细胞进行细胞悬液培养后,测其DNA经紫外线照射后的修复能力,结果证明,DNA损伤修复能力随年龄增长而降低,但牛磺酸组在各年龄段的修复能力均显著高于对照组(P<0.01),说明牛磺酸在体内具有明显的抗氧化和延缓衰老的功能,并可能作用于DNA分子水平。

Abstract:ice were used as the model of our experiment.After weanling and un-der same feeding,2 groups of mice were given ordinary water and 1% taurine(TAU)respectively.Mice were killed respectively at the age of 36 days(the early-aged stage)and 110 days(the middle-aged stage).The SOD activity,contents of MDA and lipofuscin of the liver and brain homogena-te were measured.The results showed that the SOD activity of the liver and brain homogenate in the TAU group increased obviously at the early-aged(P<0.05)and more obviously at the middle-aged(P<0.01).The content of lipofuscin and MDA in the brain homogenate had no change at the early-aged of the two groups,but they were much lower in the TAU group than those of the control group at the middle-aged(P<0.01).The same pheno,mena occurred in the content of lipofuscin of liver homogenate,but MDA had no obvious change at different age.According to the current consensus that the oxidation operates at DNA level,the mice of 110,150 and 180 days were selected for spleen suspension culture.With the same culture solution,240μmol/L TAU were added to the trial group.After irradiation with UV, ̄3H-TdR incorporation test was carried out to measure the ability of DNA repair,which displayed that DNA repair ability decreased with age,but DNA repair ability was obviously higher in TAU group than that of the control group(P<0.01),suggesting that tau has the effect of antioxidation,delays aging and acts on DNA level as well.
