摘    要:目的研究低钙对血清补体C3的影响及其可能的机制。方法雄性Wistar大鼠30只,按体重随机分为低钙组(喂饲0.15%Ca)和正常钙对照组(喂饲0.50%Ca),每组15只。喂养12w后,麻醉处死大鼠,取组织和血清。采用双向电泳技术分离大鼠血清,联合4700串联飞行时间质谱仪和蛋白质数据库分析鉴定差异表达蛋白质,并检测各组大鼠血清补体C3等生化指标。为通过人群进一步验证,在性别、年龄匹配的基础上,调查(1~3岁)佝偻病儿童及健康对照儿童,检测血清补体C3等生化指标。用real time PCR技术检测体外低钙培养的HepG2细胞及各组大鼠肝脏补体C3、furin酶mRNA表达情况。结果通过血清蛋白质组学技术筛选出低钙动物模型血清补体C3显著降低。与正常钙对照组(0.30±0.02)g/L比,低钙组大鼠血清补体C3(0.24±0.04)g/L显著降低(P<0.05)。共调查123名(1~3岁)佝偻病儿童和114名健康对照儿童,与健康对照儿童(1.52±0.10)g/L相比,佝偻病儿童血清补体C3(1.27±0.10)g/L显著降低(P<0.05)。低钙时HepG2细胞和大鼠肝脏补体C3、furin酶mRNA表达下降。结论低钙可能通过降低furin酶mRNA的表达,导致佝偻病儿童血清补体C3水平降低,机体免疫力下降,增加上呼吸道感染的危险。营养学报,2012,34(6):586-590]

Abstract:Objective To study the effect of low calcium on serum complement C3 and explore the possible mechanism.Method Thirty male Wistar rats were divided randomly into low calcium(0.15%Ca) and normal calcium(0.50% Ca) group(n=15).After 12 w,all rats were sacrificed and all samples were collected.Serum proteins were separated by 2D gel electrophoresis and differential expressed proteins were identified by 4700 Proteomics Analyzer(TOF/TOFTM) and database analysis.On the basis of gender and age-matched,serum complement C3 were tested.In(1-3y) infants with rickets and healthy infants the level of complement C3,furin mRNA expression of HepG2 cells in vitro in low calcium environment and in liver were measured using real-time quantitative PCR method.Results In low calcium animal models,the serum complement C3 was screened out to be significantly reduced by serum proteomics technology.Compared with that of normal calcium group(0.30±0.02)g/L,the concentration of serum complement C3 of low calcium rats(0.24±0.04g) g/L was significantly reduced(P<0.05).Infants with rickets(n=123,1-3 yr) and healthy infants(n=114) were investigated.Compared with that of health infants(1.52±0.10) g/L,the concentration of serum complement C3 of infants with rickets(1.27±0.10) g/L was significantly reduced(P<0.05).The expression of complement C3,furin mRNA in both HepG2 cells and liver was decreased under low calcium intake.Conclusion Low calcium may decrease complement C3 by reducing the level of furin mRNA expression and this may be the reason for increasing risk of respiratory infections when immunocompromised.ACTA NUTRIMENTA SINICA,2012,34(6):586-590]
