低氧对RAW264.7细胞中糖皮质激素受体表达的调节

摘    要:目的观察小鼠巨噬细胞系RAW264.7细胞在低氧培养箱(1%O2)内不同时间段糖皮质激素受体(glucocorticoid receptor,GR)mRNA和蛋白表达的变化.探讨低氧对RAW264.7细胞中GR表达的调节。方法采用体外培养的小鼠巨噬细胞系RAW264.7细胞,将细胞接种于60mm dish内.细胞长至50%时,将培养液置换为含5%无激素胎牛血清的无糖DMEM培养液,放入低氧培养箱内,同时输入含1%O2,5%CO2,94%N:的混合气体,分别于低氧处理4、8、12h后将细胞取出,抽提RNA或者蛋白.应用real—timePCR和Western印迹分析测定GRmRNA和蛋白的表达水平.以未低氧处理RAW264.7细胞为对照。结果低氧处理4h可使RAW264.7细胞中GRmRNA的表达明显上调为对照的1.8倍(P〈0.05),处理至12hGRmRNA的表达仍高于对照,约为对照的2.7倍(P〈0.05);低氧处理RAW264.7细胞8hGR蛋白的表达也明显升高,约为对照的1.7倍(P〈0.05),至低氧处理12h,GR蛋白的表达仍高于对照。结论低氧可以上调体外RAW264.7细胞GRmRNA和蛋白的表达。

Abstract:Objective To investigate the changes of the glucocorticoid receptor in vitro induced by hypoxia for 12h. Methods RAW264.7 cells were treated by hypoxic conditions for 12h for detection of GR mRNA and protein level by real-time PCR and western blot respectively.
