超微六味地黄汤对孕期被动吸烟胎鼠脑Bax、Bcl-2和Casepase-3的影响

摘    要:目的研究六味地黄汤超微和传统饮片对胎鼠脑发育不全的干预作用,并从细胞凋亡角度探讨其机理。方法采用孕鼠被动吸烟造成胎鼠脑发育不全,分别给予六味地黄汤超微和传统饮片进行治疗。采用免疫组化法检测Bax、Bcl-2和Casepas’e-3。结果模型组三项指标均明显高于正常组(P〈0.01);治疗组与模型组相比Bax和Caspase-3明显减少(P〈0.01);但仍高于正常组(P〈0.01);治疗组与模型组相比Bcl-2变化不大(P〉0.05)。超微全量组和1/3量组Caspase-3表达较传统组少(分别为P〈0.05和P〈0.01);超微全量组Caspase-3的表达较超微1/3量组少(P〈0.01)。结论孕期被动吸烟可以导致胎鼠脑Bax、Bcl-2和Casepase-3表达水平升高,六味地黄汤能降低Bax和Casepase-3的表达水平。

Abstract:Objective To study the effects of Liuweidihuang Tang superfine-powder and traditional decoction on mouse embryo atelencephalia by passive smoking in pregnancy,and to study its mechanism through cell apoptosis.Methods Making the mouse embryo atelencephalia model by passive smoking in pregnancy,we administrated Liuweidihuang Tang superfine-powder and traditional decoction,and measured the changes of Bax,Bcl-2 and Casepase-3.Results There was a significant increase of embryo brain Bax,Bcl-2,and Casepase-3 in model group as compared with control group(P<0.01).There was a significant decrease of Bax and Casepase-3 in treatment groups as compared with model group(P<0.01),but the expressions of Bax and Casepase-3 were still higher than those of control group(P<0.01).No significant change of Bcl-2 was found in treatment groups as compared with model group(P>0.05).The expression of Caspase-3 in superfine-powder group and 1/3 superfine-powder group was less than that of traditional group(P<0.05,P<0.01).The expression of Caspase-3 in superfine-powder group was less than that of 1/3 superfine-powder group(P<0.01).Conclusions The expressions of Bax,Bcl-2 and Casepase-3 are increased on mouse embryo by passive smoking in pregnancy,and Liuweidihuang Tang can inhibit the changes of Bax and Casepase-3.
