摘    要:目的 探讨枸杞多糖(LBP)对创伤后应激障碍(PTSD)大鼠抑郁行为的影响及作用机制。方法 SD大鼠随机分为对照组、模型组、帕罗西汀组和LBP组,每组12只。除对照组外,其余各组大鼠均采用单一连续应激方法制备PTSD动物模型。造模后d1,药物处理组分别灌胃给予盐酸帕罗西汀(4.2 mg/kg)和LBP(25、50、100 mg/kg),1次/d,连续3周。对照组和模型组灌胃等量生理盐水。蔗糖消耗实验和强迫游泳实验检测大鼠的行为变化;免疫组织实验检测大鼠N - 甲基 - D - 天门冬氨酸受体 2B亚基(NR2B)和钙调蛋白激酶 Ⅱ(CaMK Ⅱ )蛋白的表达情况;ELISA法检测血清皮质酮浓度。结果 与对照组相比,模型组大鼠糖水消耗率下降(P<0.05),强迫游泳不动时间延长(P<0.05),前额叶皮质NR2B和CaMK Ⅱ蛋白的表达量升高(P<0.05),血清皮质酮浓度升高(P<0.05)。LBP可以改善上述指标变化,具有统计学意义(P<0.05)。结论 LBP可以通过下调前额叶皮质NR2B - CaMK Ⅱ信号通路、降低血清皮质酮浓度,改善PTSD大鼠的抑郁行为。

Abstract:Objective To explore the effect and mechanism of lycium barbarum polysaccharide (LBP) on depression behavior of rats with post-traumatic stress disorder(PTSD). Methods Sprague-Dawley rats were randomly divided into control group, model group, paroxetine group and LBP group, with 12 rats in each group. All of the rats except those in control group were treated with single prolonged stress to establish PTSD model. The first day after modeling, rats in paroxetine group and LBP groups were ig given paroxetine hydrochloride (4.2 mg/kg) or LBP (25 mg/kg, 50 mg/kg, 100 mg/kg), once a day, for a total of 3 weeks. Rats in control group and model group were accepted the same dose of saline. Sugar preference test and forced swimming test were carried out to detect the behavioral changes. Immunofluorescence technique was used to detect the expression of NR2B and CaMK Ⅱ in prefrontal cortex. The level of serum corticosterone was measured by ELISA. Results Compared with control group, the sugar consumption rate was decreased in model group(P<0.05), the immobility time was extended(P<0.05), the expressions of NR2B and CaMK Ⅱ were increased(P<0.05). Meanwhile, the level of serum corticosterone was tangibly ascended. LBP treatment reversed all the above indicators(P<0.05). Conclusion LBP could improve the depression-like behavior with PTSD rats, which might be related with lowering NR2B-CaMK Ⅱ signaling pathway and reducing serum cortisol.
