摘    要:目的探讨VEGF-C反义寡核苷酸(VEGF-C ASODN)和姜黄素联合化疗对人卵巢癌细胞SKOV3生长粘附侵袭能力的抑制作用。方法 MTT法检测VEGF-C ASODN和姜黄素联合化疗对卵巢癌细胞的增殖抑制效应;MTT法检测卵巢癌细胞与细胞外基质粘附能力,体外侵袭实验检测卵巢癌细胞侵袭人工基底膜的能力。结果 VEGF-C ASODN和姜黄素联合化疗对卵巢癌细胞有明显的增殖抑制作用,使卵巢癌细胞对细胞外基质粘附率明显降低,穿透重组基质膜数目明显下降。结论 VEGF-C ASODN和姜黄素联合化疗能明显抑制卵巢癌细胞体外实验中的生长粘附侵袭能力,可望为卵巢癌提供一种有效的联合化疗方法。

Abstract:Objective To investigate the inhibition of VEGF-C antisense oligodeoxynucleotide(ASODN) and curcumin combination chemotherapy on the proliferation and adhesion and invasion of human ovary cancer cell line SKOV3 in vitro.Methods The ovary cancer cell proliferation in VEGF-C ASODN and curcumin combination chemotherapy was detected by MTT.The adhesion to artifical basement membrane Matrigel was measured by MTT assay.The invasion was evaluated by Tran swell chambers attached with polycarbonate filters and reconstituted basement membrane(Matrigel).Results VEGF-C ASODN and curcumin combination chemotherapy could significantly inhibit the growth of SKOV3 cells.The adhesive and invasive cell numbers decreased markedly.Conclusion VEGF-C ASODN and curcumin combination chemotherapy could inhibit the proliferation and adhesion and invasion of SKOV3 cells remarkably and could provide effective combination chemotherapy for ovarian cancer.
