重组人生长激素对内毒素诱导人巨噬细胞产生TNF-α、IL-1β含量的影响

摘    要:目的研究重组人生长激素(rhGH)对内毒素(LPS)诱导巨噬细胞产生炎性因子的影响。方法培养人单核巨噬细胞株U937细胞,体外经PMA(phorbol 12-myristate 13-acetate)诱导成熟后,设为5组:空白对照组、LPS组(给予40μg/mL LPS)、rhGH低剂量组(给予2U/L rhGH+40μg/mL LPS)、rhGH中剂量组(给予4U/L rhGH+40μg/mL LPS)及rhGH高剂量组(给予8U/L rhGH+40μg/mL LPS),采用酶联免疫吸附试验(ELISA)测定各组细胞上清液中炎性因子肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1β的含量。结果经PMA分化成熟后的U937细胞在LPS诱导下,TNF-α、IL-1β的含量明显增加,LPS组与空白对照组比较,差异有显著性(P〈0.05);rhGH各组TNF-α、IL-1β的含量则明显下降,与LPS组比较,差异有显著性(P〈0.05)。结论rhGH可抑制LPS刺激巨噬细胞分泌炎性因子TNF-α、IL-1β,故rhGH对LPS所致疾病如脓毒血症等可能有潜在的治疗作用。

Abstract:Objective To investigate the effect of recombinant human growth hormone (rhGH) on the inflammatory cytokine secretion from maerophage stimulated by lipopolysaccharide (LPS). Methods Human mononuclear macrophage U937 cells were cultured and induced to be mature by phorbol 12-myristate 13-acetate in vitro, then U937 cells were divided into five groups: control group, LPS group (40/μg/mL LPS included), low dose rhGH group (2U/L rhGH + 40μg/mL LPS included), middle dose rhGH group (4U/L rhGH + 40μg/mL LPS included) and high dose rhGH group (8U/L rhGH + 40μg/mL LPS included). TNF-α and IL-1β released from the U937 cells were detected by ELISA. Results The concentration of TNF-α and IL-1β released from the stimulated U937 cells in LPS group increased significantly than that in the controls (P〈0. 05) ; and the levels of TNF-α and IL-1β in all the rhGH groups were significantly lower than those in LPS group (P〈0. 05). Conclusion rhGH can inhibit the secretion of TNF-α and IL-1β from macrophage stimulated by LPS, so rhGH may have potential curative effect on diseases induced by LPS, such as sepsis.
