汉坦病毒单克隆抗体的制备和鉴定

本文用汉坦病毒A16和R22株活毒免疫BALB／c小鼠，取牌细胞与sp2／0骨髓瘤细胞融合，制备了6株分泌抗汉坦病毒单克隆抗体的杂交癌细胞系。单克隆抗体IgG亚类鉴定表明，其中2株为IgG1，1株为IgG2b，3株为IgG2a。用间接免疫荧光检测单抗腹水滴度，分别在1：10240～1：81920之间；用反向被动血凝抑制试验测定单抗腹水，滴度均低于1：20。免疫印迹试验表明，6株单克隆抗体均为抗核蛋白单抗。对已知型别病毒的反应话表明，其中3株为组特异性单抗，1株为汉滩型特异性单抗，2株为汉城型特异性单抗。还用6株单抗对宁夏新分离毒株进行抗原性分析。

Preparation and Characterization of Monoclonal Antibodies against Hantavirus

Six hybridoma cell lines producing monoclonal antitalies to hantavir-us were established by fusion of sp2/0 mice rnyeloma cells with spleen cells of BALB/c mice immunized with Hantavirus strain 76- 118 and R22. Among these, two were of the IgGl, one was of IgG2b and three were of IgG2a.Ascitic titres of the antibodies ranged from 1: 10 240 to 1: 81 920 when tested by indirect immunofiuorescent antibody staining rnethod. Reversed passive hemagglutination inhibition test dernonstrated that the hemagglutination inhibition titres of the six antibodies were lower than l: 20. Western-blotting assay showed the six antitheies were all binding to the nucleocapsid protein. According to cross-reactivities with other hantavirus, the antibodies were classified into three groups: 3 antitheies cross-reacting with Hantaan and Seoul virus strains, l specific to Hantaan serotype and 2 specific to seul serotyPe. 6 antibodies were also used in antigenic analysis of virus strains isolated from Ningxia.
