脉冲场凝胶电泳对肠球菌的分子检测与同源性分析

摘    要:目的对40株肠球菌进行分子同源性检测分析,探讨脉冲场凝胶电泳(pulsed field gel electrothoresis, PFGE)在分子流行病学方面的价值. 方法应用PFGE对40株分离自住院患者的肠球菌进行同源性分析. 结果 40株肠球菌经脉冲场凝胶电泳在30~500 kb之间显示12~18条大小不同的DNA片段;40株肠球菌表现出大小不一的同源性:3对肠球菌100%同源,来自同一病房的4对肠球菌分别为78.27%、72.73%、72.73%、80.01%同源;来自不同病房两对肠球菌分别为80.01%、77.78%同源,其余菌株则显示了较低的同源性或者无同源性. 结论 40株肠球菌未见同一克隆的暴发流行;PFGE是细菌分子流行病学分析的理想方法.

Abstract:OBJECTIVE To analyze the molecular homology in Enterococcus and the significance of pulsed field gel electrophoresis (PFGE) in molecular epidemiology. METHODS PFGE was used to analyze the homology of 40 Enterococcus strains isolated from inpatients. RESULTS DNA bands of different size from 30-500 kb were present in the gel, different homology was shown among the 40 strains. Three couples were 100% similar, 4 couples of strains from their same wards were of the homology of 78.27%, 72.73%, 72.73%, and 80.01%, respectively, 2 couplesfrom their different wards were 80.01%, 77.78% of homology, respectively, others were either of poor homology or no homology. CONCLUSIONS No clonal ourbreak is seen among the 40 Enterococcus strains; PFGE is a discriminatory and reproducible strain typing technique in epidemiologic analysis of microorganisms.
