吞噬细胞吞噬作用实验方法的改进

目的探讨和改进实验方法,以便更清晰、有效地观察吞噬细胞对外来异物的吞噬作用。方法小吞噬试验是用金黄色葡萄球菌感染昆明小鼠、大吞噬试验是将鸡红细胞注入Hartly豚鼠体内,然后取鼠腹腔液涂片、甲醇固定,分别用瑞氏染色法和瑞氏-姬姆萨染色法进行染色,镜下观察吞噬细胞对外来异物的吞噬作用。结果瑞氏-姬姆萨染色法制作的标本比瑞氏染色法制作的标本显微镜下观察吞噬细胞的形态结构更清晰,能更好地显示出不同种类的细胞及细胞的不同成分的色彩特征,可以更快、更有效地分辨各类细胞的细胞核、细胞浆和被吞噬的外来异物。结论采用瑞氏-姬姆萨染色法制作的吞噬作用实验标本在镜下形态结构清晰、色彩特征明显,易于分辨细胞成分和外来异物的种类,便于学生观察和计数,有助于学生对吞噬细胞吞噬作用的理解和掌握。

Experiment Improvement for the Phagocytosis of Phagocytes

Objective To improve experimental methods in the phagocytosis of phagocytes.Methods K.M mice were infected with S.aureus and HartlyCavia Porcelluswas injected with red blood cell of chicken respectively.Then the celiac liquid was smeared and fixed on the slides,whichwere stainedwithWright’s staining andWright-Giemsa staining to observe the resultswith themicroscope.Results Wright-Giemsa staining was better than Wright’s staining based on the morphological structure of phagocytes.Cell nucleus,plasma and phagocytosed foreign matter could be distinguished more quickly,clear and effectively.Conclusions By using Wright-Giemsa staining,clear morphological structure and obvious colour characteristics can be observed.This method is useful for students fully understanding the phagocytosis.
