VEGF-C及癌周淋巴管密度与头颈鳞癌颈部淋巴结转移的关系

摘    要:目的探讨血管内皮细胞生长因子-C(VEGF-C)表达及癌周淋巴管密度(P-LVD)与头颈鳞癌(HNSCC)颈部淋巴结转移的关系。方法采用免疫组化法,检测104例HNSCC组织中VEGF-C及癌周LYVE-1的表达,幷计算P-LVD,分析P-LVD及VEGF-C表达与其临床病理因素的关系。结果 104例HNSCC组织中,VEGF-C阳性表达率显著高于正常头颈部黏膜组织(74.04%vs 0,P〈0.05)。淋巴结转移组VEGF-C阳性表达率高于无淋巴结转移组(P〈0.05)。对LYVE-1标记的淋巴管密度(LVD)分析显示,癌周组织P-LVD高于癌内及正常组织(P〈0.05),而VEGF-C阳性表达者及颈部淋巴结转移者P-LVD分别显著高于VEGF-C阴性表达者和无颈部淋巴结转移者(P〈0.05)。结论 HNSCC中VEGF-C表达及P-LVD与肿瘤淋巴结转移密切相关,对判断肿瘤侵袭、转移具有一定参考价值。

Correlation of VEGF-C Expression and Peritumoral Lymphatic Vessel Density with Nodal Metastasis in HNSCC

Abstract:Objective To investigate the relationship between VEGF-C expression,peritumoral lymphatic vessel density(P-LVD) and lymph node metastasis in head neck squmaous cell carcinoma(HNSCC).Methods P-LVD and expression of VEGF-C was detected,respectively,by immunohistochemical staining with the antibody to VEGF-C and LYVE-1 in 104 cases of HNSCC.The correlation of clinico-pathological feature with VEGF-C expression and PLVD wes evaluated.Results The positive rates of VEGF-C in HNSCC tissues and normal oral mucousa membrane were 74.04% and 0 respectively(P0.05).The expressions of VEGF-C was significantly higher in lymph node metastasis positive patients than those of metastasis negative patients(P0.05).Analysis of LVD labelled by LYVE-1 showed that P-LVD was significantly higher than intratumoral LVD(I-LVD) and LVD in normal tussues(P0.05).Patients of VEGF-C positive group and lymph node metastasis positive group had,respectively,higher P-LVD than those of VEGF-C negative group and lymph node metastasis negative group(P0.05).Conclusion VEGF-C expression and P-LVD were closely correlated with regional lymph node metastasis in HNSCC and may be valuable in the evaluation of potential for invasivness and metastasis.
