调气消积汤对人肺腺癌A549细胞肿瘤相关基因表达的影响

目的:检测调气消积汤对人肺腺癌A549细胞周期及肿瘤相关基因IL-8、S28、Sp1表达的影响，探讨该方抗肺癌的分子机制。方法:实验共分为5组:空白对照组、调气消积汤低、中、高剂量组（浓度分别为5%、15%、20%）及顺铂组，常规方法制备含药血清。以不同剂量的调气消积汤及顺铂制作含药血清并分别作用于A549细胞，用流式细胞仪检测细胞周期，采用RT-PCR法检测各组IL-8、S28、Sp1 mRNA的表达水平。结果:随着药物浓度的增加，调气消积汤各剂量组的G0/G1期细胞比例逐渐降低，S期细胞比例逐渐升高（P<0.01）。调气消积汤各剂量组及顺铂组IL-8、S28、Sp1 mRNA表达水平明显低于空白对照组，差异有统计学意义(P<0.05 或 P<0.01）。调气消积汤高剂量组与顺铂组比较，差异无统计学意义（P>0.05）。结论:调气消积汤可以阻止肿瘤细胞进入增殖周期；下调相关基因IL-8、S28、Sp1 mRNA的表达水平，可能是调气消积汤发挥抑瘤作用的分子机制之一。

The effect of tumor -associated genes expression of human lung adenocarcinoma A549 cells treated with Tiaoqixiaoji Decoction

Objective:To approach the molecular mechanism of Tiaoqixiaoji Ddecoction(TQXJ)on tumor -asso-ciated genes expression of IL -8,S28,Sp1 of human lung adenocarcinoma A549 cells.Methods:Cells were divided into 5 groups:Blank group,TQXJ low,median,high dose groups(concentrations were 5%,15%,20%)and DDP group,regular prepare concentrations of drug serum.A549 cells after treatment with different concentrations of Tiao-qixiaoji Decoction serum were observed.The flow cytometry was used to determine the cell cycle,and the genes ex-pression of IL -8,S28,Sp1 mRNA were detected by RT -PCR method.Results:With the increase of drug concentra-tion,the proportion of cells in G0 /G1 phase was decreased,and that in S phase was increased for the each dose TQXJ groups than that of the blank group(P <0.01).Each dose TQXJ group and DDP group all could reduce the expres-sion of IL -8,S28,Sp 1 mRNA,there were statistical difference compared with the blank group (P <0.05 or P <0.01).Compared with high dose TQXJ group and the DDP group,there was no statistical difference between them(P>0.05).Conclusion:The TQXJ may block the cancer cells entering the proliferative phase,and it's activity of inhibi-ting the growth of tumor may be relative to decreasing the expression of IL -8,S28,Sp1 mRNA.
