长链非编码RNA CASC9在胃癌组织中的表达及其临床意义

目的:检测长链非编码RNA CASC9（cancer susceptibility candidate 9）在胃癌组织和细胞中的表达，探讨CASC9与胃癌患者临床病理特征及预后的关系。方法:利用RT-PCR检测胃癌组织、细胞及配对癌旁组织、正常胃黏膜细胞中CASC9的表达；利用单因素方差分析CASC9与胃癌患者临床病理特征之间的关系；利用生存曲线分析CASC9与胃癌患者术后5年生存率的关系；利用COX风险比例模型分析CASC9与胃癌患者预后的关系。结果:CASC9在胃癌组织、细胞中表达水平高于癌旁组织和正常胃黏膜细胞；CASC9的表达与胃癌的Bormann分型及TNM分期相关（P＜0.05）；CASC9高表达的胃癌患者术后5年生存率低于低表达的患者，CASC9高表达是胃癌患者独立的预后因素。结论:CASC9在胃癌中表达升高，并与胃癌患者生存及预后密切相关。

Expression and clinical significance of long non-coding RNA CASC9 in gastric cancer tissues

Objective:To investigate the expression level of long non-coding RNA (lncRNA) cancer susceptibility candidate 9 (CASC9) in gastric cancer,and evaluate its prognostic value in gastric cancer.Methods:Gastric cancer cells and normal gastric mucosal cells were tested in this study.Cancer tissue as well as adjacent tissue from patients diagnosed with gastric cancer were also harvested in this study.Expression of CASC9 was measured by real-time PCR.ANOVA analysis was performed to evaluate the association between CASC9 and clinical characteristics of gastric cancer patients.K-M survival analysis was utilized to determine correlation of CASC9 expression with 5-year survival.COX's proportional hazards regression models were applied to analyze the relationship between CASC9 and prognosis of gastric cancer patients.Results:CASC9 was upregulated in gastric cancer tissues and cells compared with adjacent tissues and normal gastric mucosal cells.Expression of CASC9 was closely associated with Bormann type and TNM stage of gastric cancer (P＜0.05).Expression of CASC9 was negatively correlated with 5-year survival rate.CASC9 was an independent prognostic risk factor of gastric cancer.Conclusion:lncRNA CASC9 is upregulated in gastric cancer and is negatively associated with survival outcome of gastric cancer.
