大黄素对人肺腺癌A-549细胞增殖周期影响的实验研究

目的：观察大黄素对人肺腺癌A-549细胞增殖及周期的影响。方法：设计空白对照组及不同浓度的大黄素溶液作用于A-549细胞，MTT比色法测定A-549细胞生长的抑制率；流式细胞仪分析A-549细胞增殖周期的变化。结果：大黄素对人肺腺癌A-549细胞有明显的生长抑制作用，其抑制作用呈浓度依赖性，其抑制率随药物浓度升高而增加。400mg／L时达到83．33％，实验组与对照组之间差异有统计学意义，P=0．000；对A549细胞具有细胞周期阻滞作用，可将其阻滞于G0／G1期，阻止细胞进入S期。结论：大黄素对人肺腺癌A549细胞的增殖生长具有显著的抑制作用；并可使A-549细胞增殖周期停滞于G0／G1期，而阻止其进入S期。

Study on the effect of emodin on the cell proliferation periods of humanlung adenocarcinoma cell line A-549

OBJECTIVE: To investigate the effect of emodin on the cell proliferation periods of human lung adenocarcinoma cell line A-549. METHODS: A-549 cells were divided into control group and emodin groups. The growth restrain rate of the A-549 cells was determined by MTT, and the cell proliferation periods of the A-549 cells were analysed. RESULTS: Emodin had obviously the function of the restrain of the A-549 cell growth,the function intension dependented on the density of emodin, the growth restrain rate of the A-549 cells was along with the density of emodin, and it was 83.33% when density of emodin was 400 mg/L. Through repeate measure analysis of variance, it was found that there was a significant difference between the experiment groups and control group, P=0.000. Emodin had the function of block cell peridos on A-549, it stoped the A-549 cells at G0/G1 period. CONCLUSION: Emodin has obviously the function of the restrain of the A-549 cell growth, but also has the function of the block cell peridos on A-549, it can stop A-549 cells at G0/G1 period.
