摘    要:目的:研究CPT-11对食管癌细胞株ECa109细胞周期及其放射敏感性的影响。方法:将ECa-109细胞株分为单纯培养组(对照组)、单纯化疗组、单纯放疗组、同时化放疗及时相化放疗组;MTT法检测细胞存活率,选择CPT-11的最佳实验浓度;流式细胞仪(FCM)观察CPT—11诱导ECa—109细胞阻滞于G2期的时间,作为时相化放疗组加药后行放疗的时间点,并分析各组细胞的凋亡率;克隆形成实验观察CPT-11作用后各放疗组细胞的存活率。结果:3.85μg/mL CPT-11为最佳实验浓度;FCM检测细胞周期显示,细胞处于G2期的时间点约为加药后12h;FCM检测细胞凋亡显示,化疗组、放疗组、同时化放疗组、时相化放疗组疗效依次递增P〈0.05;化疗组与对照组差异无统计学意义,P〉0.05;细胞克隆形成实验显示,CPT-11能降低ECa-109放疗后存活率;时相化放疗组及同时化放疗组均优于单纯放疗组,且前者优于后者;两者的放疗增敏比分别为1.39和1.11。结论:CPT11对ECa109细胞株有放射增敏作用且时相化放疗的增敏作用更强。

Effection of cell cycle and radiosensitivity of OPT-11 on Esophageal cancer cell line-109

Abstract:OBJECTIVE:To explore the effection of cell cycle and radiosensitivity of CPT -11 on Eea-109 cell line. METHODS: There were five groups including group of culture (contrast group), group of chemotherapy,group of radiotherapy, group of homochronous ebemo radiotherapy and group of phase ehemoradiotherapy in the test. Cell survival rate was detected by MTT in order to chose the best practical density of CPT-11. The time that CPT -11 induced ECa-109 to block at G2 stage was detected by flow cytometry, which could determine time point of radiotherapy after CPT-11 added in phase chemoradiotherapy. Cell survival rate after CPT -11 added in each group of radiotherapy was detected by experiment of cell clone formation. RESULTS: The best practical density of CPT- 11 was 3. 85 μg/mL. Twelve hours after CPT-11 added was the time point which CPT- 11 induced ECaq09 to block at G2 stage. The effect increased progressively one by one on the group of chemotherapy,group of radiotherapy, group of homochronous chemoradiotherapy and group of phase chemoradiotherapy phase chemoradiotherapy (P 〈 0.05 ). There weren't difference between the group of chemotherapy and contrast group (P2〉0.05). Postradiotherapy CPT -11 could decrease survival rate in Eca 109 cell line. The effect on group of phase chemoradiotherapy and homochronous chemoradiotherapy were higher than that of radiotherapy, moreover, the effect on the former was higher than the latter; The ratio of radiosenitivity of the two groups respectively were 1. 39 and 1. 11 in the experiment of cell clone formation. CONCLUSION: CPT-11 can increase radio sensitivity in Eca 109 cell line and can increase more highly by phase chemoradio therapy as well.
