胚胎小鼠肾脏发育过程中eNOS表达与细胞凋亡的关系

摘    要:目的观察胚胎小鼠不同发育阶段肾组织中内皮型一氧化氮合酶(eNOS)的表达和细胞凋亡的特征,探讨eNOS与细胞凋亡的关系。方法应用原位末端标记(TUNEL)法检测胚胎小鼠肾组织中的细胞凋亡,免疫组织化学方法和体视学方法检测eNOS的表达。结果不同发育阶段的肾组织中均可见到TUNEL染色阳性的细胞,凋亡指数从E14d开始增高,E18d达到高峰。eNOS表达于皮质生肾区、近端小管和远端小管,表达量随着发育逐渐增加。结论细胞凋亡和eNOS的表达在小鼠肾脏胚胎发育过程中具有重要的作用,细胞凋亡与eNOS的表达有一定的相关性。

Abstract:Objective To study the expression of endothelial nitric oxide synthase (eNOS) and apoptosis during the development of mouse embryonic kidney, and to explore the relationship between eNOS expression and apoptosis during the developmental process. Methods The apoptosis was determined by Tdtmediated dUTP nick end labelling (TU NEL). The expression of eNOS was examined by immunohistochemistry and stereology. Results TUNEL-positive cells could be easily observed at all stages of developing kidneys. The number of apoptotic cells increased from E14d and peaked at E18d. The eNOS was experessed in nephrogenic zone, proximal tubules and distal tubules, increasing along with the development. Conclusion The expression of eNOS and apoptosis play an important role in the development of mouse embryonic kidney. There is a relationship between the eNOS expression and apoptosis.
