微小RNA-138-5p在前列腺癌细胞中的表达及对其侵袭能力的影响

摘    要:目的探讨微小RNA(miR)-138-5p在前列腺癌(PCa)中的表达及对前列腺癌侵袭、迁移、凋亡等能力的影响。方法miR-138-5p模拟物转染正常前列腺上皮细胞(RWPE)-1、DU145细胞,实时定量反转录聚合酶链反应(RT-qPCR)用于检测转染前后细胞miR-138-5p的表达。细胞计数试剂盒(CCK-8)实验、Transwell实验、划痕实验和凋亡实验分析miR-138-5p对PCa侵袭、迁移等能力的影响。GraphPad Prism 6 V6.01用于数据分析,组间比较采用Student’s t检验。结果RT-qPCR结果表明miR-138-5p在DU145细胞中的表达量(0.470±0.111)低于RWPE-1细胞(5.120±0.824);转染后,RWPE-1 mimic中miR-138-5p的表达量(18.150±2.742)明显高于RWPE-1 NC(4.930±0.586,t=6.670,P<0.01),DU145 mimic中miR-138-5p的表达量(9.865±1.006)高于DU145 NC(0.526±0.145,t=12.990,P<0.01),差异均有统计学意义;提高miR-138-5p表达水平后,PCa细胞活力下降;DU145 mimic的迁移面积抑制率(1.50±0.08)%]高于NC组(1.00±0.03)%,t=8.382,P<0.01],差异有统计学意义;DU145 mimic的侵袭细胞数(102.00±5.96)个]低于NC组(198.00±15.09)个,t=8.392,P<0.01],差异有统计学意义;DU145细胞mimic组的凋亡率(17.660±1.312)%]高于NC组(4.990±0.674)%,t=12.150,P<0.01],差异有统计学意义,PCa细胞的凋亡增加。结论miR-138-5p可以负向调节PCa细胞的活力、迁移和侵袭,促进PCa细胞凋亡.

Abstract:Objective To explore the expression of microRNA(miR)-138-5p in prostate cancer(PCa)and its effects on the invasion,migration and apoptosis of PCa.Methods Normal prostate epithelial cells(RWPE-1)and DU145 cells were transfected after cell culture.The quantitative real-time polymerase chain reaction(RT-qPCR)was used to detect the expression of miR-138-5p before and after transfection.The effects of miR-138-5p on invasion and migration of PCa were analyzed by cell counting kit-8(CCK-8)assay,Transwell assay,Wound-healing assay and apoptosis test.GraphPad Prism 6 V6.01 was used for data analysis and Student’s t test was used to assess statistical significance between groups.Results RT-qPCR showed that the expression of miR-138-5p in DU145 cells(0.470±0.111)was lower than that in RWPE-1 cells(5.120±0.824).After transfection,the expression of miR-138-5p in RWPE-1 mimic group(18.150±2.742)was significantly higher than that in RWPE-1 normal control group(4.930±0.586,t=6.670,P<0.01),and that in DU145 mimic group(9.865±1.006)was higher than that in DU145 normal control group(0.526±0.145,t=12.990,P<0.01).After increasing the expression level of miR-138-5p,the viability of PCa cells decreased.The inhibition rate of migration area in DU145 mimic group(1.50±0.08)%]was higher than that in normal control group(1.00±0.03)%,t=8.382,P<0.01].The number of invasive cells in DU145 mimic group(102.00±5.96)was less than that in normal control group(198.00±15.09,t=8.392,P<0.01).The apoptosis rate of DU145 cells in mimic group(17.660±1.312)%]was higher than that in normal control group(4.990±0.674)%,t=12.150,P<0.01],and the apoptosis of PCa cells increased.Conclusion MiR-138-5p can negatively regulate the viability,migration and invasion of PCa cells and promote the apoptosis of PCa cells.
