E2F1对H2O2诱导的人乳腺癌细胞凋亡的影响

摘    要:目的 观察E2F1对H2O2诱导的人乳腺癌细胞MDA-MB435凋亡的影响.方法 通过脂质体法转染目的质粒pcDNA3.1(+)/E2F1进人人乳腺癌细胞株MDA-MB435,筛选稳定表达E2F1的细胞克隆,通过Western blot法鉴定高表达E2F-1的MB435细胞克隆C1、C2和C3,并应用锥虫蓝染色法检测100 μmol/L H2O2处理后细胞的存活率变化.结果 在细胞克隆MB435/E2F1-C1、C2和C3中有高水平的E2F1表达,这些高表达E2F1的肿瘤细胞经100 μmol/L H2O2刺激后细胞存活率(分别为63.4%、54.2%和32.9%)相对于对照组(90.9%)均明显降低,且其降低的水平与E2F1的表达水平成正比,差异有统计学意义(P<0.01).结论 E2F1对H2O2诱导的人乳腺癌细胞凋亡有促进作用.

Abstract:Objective To study the influence of E2F1 on signaling oxidative stress-induced apoptosis of MDA-MB435 cells.Methods pcDNA3.1 ( + )/E2F1 was transfected into MB435 cells by using LipofectAMINE and stable cell clones expressing high level E2F1 (MB435/E2F1-C1,C2,C3) were selected,and the expression level of E2F1 was detected by using Western blotting.E2Fl-mediated apoptosis of MDA-MB435 cells treated with 100 μmol/L H2O2 was examined by Trypan blue exclusion.Results There were high levels of E2F1 protein in the three stable clones expressing ectopic E2F1,and the viability of cells with high levels of E2F1 was greatly reduced after treatment with 100 μmol/L H2O2 as compared with the control cells (63.4%,54.2% and 32.9% vs.90.9%,P<0.01).Conclusion E2F1 can increase apoptosis of breast cancer cells in response to H2O2 treatment.
