洛伐他汀对系膜细胞凋亡及Bcl-2和Bax基因表达的影响

摘    要:目的 观察洛伐他汀对系膜细胞凋亡及Bcl-2和Bax基因表达的影响。并探讨其作用的可能机制。方法 用不同浓度的洛伐他汀处理大鼠系膜细胞,用流式细胞仪观察细胞凋亡指数,吖啶橙荧光活体染色观察细胞凋亡率,电镜观察凋亡细胞形态,免疫印迹法及细胞免疫结合图文分析观察洛伐他汀对系膜细胞Bcl-2和Bax基因表达的影响。结果 洛伐他汀对系膜细胞凋亡有明显诱导作用,呈一定剂量依赖性;并能明显抑制Bcl-2基因表达,促进Bax基因表达,致使两者比例明显下降。结论 洛伐他汀可剂量依赖性地诱导大鼠系膜细胞凋亡,其机制可能通过对凋亡抑制或促进基因的调控,改变Bcl-2/Bax比值而诱导MC的凋亡。

Abstract:Objective To investigate the effect of lovastatin on apoptosis of rat mesangial cell (MC) and expression of gene Bel-2 and Bax, and to inquire into its possible mechanism. Methods Rat mesangial cells were incubated with dillerent concentrations of lovastalin. Apoptosis indexes were assessed by How rytomelry, apoptosis rate analyzed by acridine orange immunofluorescence vital staining, morphology of upoplosis discriminated by electroscope, effect of lovastatin on expression of gene Bcl-2 and Bax investigated by Western blot and cell-mediated immunity associated with videotexl analysis. Results Lovastatin could significantly induce apoptosis of MCs, with dose-dependent correlation to some extent. Expression of gene Bcl-2 and Bax was down-regulated and up-redulated respectively, the ratio of Bcl-2 and of Bax was significantly decreased. Conclusions Lovastatin induces apoptosis of rat mesangial cells in dose-dependent manner. Its mechanism may be that apoptosis of MC results from altered ratio of Bcl-2 and Bax level by apoptotic down-regulation or up-regulalion of control gene.
