摘    要:目的探讨Ⅱ型胶原蛋白(CollagenⅡ,COL2)表达水平与骨质疏松之间的相关性。方法收集2017年1月至2019年12月在新疆维吾尔自治区人民医院骨科中心关节老年病区行手术治疗的髋部骨折60例患者,按照双能X线骨密度(DXA)检测分为骨量正常、骨量减少、骨质疏松和严重骨质疏松4组,每组15例,分别于术中取患者股外侧肌肉组织行RT-PCR及免疫组化检测COL2 mRNA及蛋白表达水平的差异性,并综合分析COL2表达水平与骨质疏松的相关性。结果 RT-PCR及免疫组化检测结果显示,骨量正常组COL2 mRNA及蛋白表达水平与骨量较少组之间差异无统计学意义(P0.05);骨量减少组COL2 mRNA及蛋白表达水平明显高于骨质疏松组,且差异有统计学意义(P0.05);骨质疏松组COL2 mRNA及蛋白表达水平明显高于严重骨质疏松组,且差异有统计学意义(P0.05)。结论 COL2表达水平与骨质疏松呈负相关,COL2可能是预防绝经后女性骨质疏松性髋部骨折的治疗靶点。

The relationship between COL2 expression and osteoporosis in postmenopausal women
Abstract:Objective To explore the correlation between type II collagen (COL2) expression level and osteoporosis. Methods Sixty patients with hip fractures who underwent surgical treatment in the Orthopedic Center of the People''s Hospital of Xinjiang Uygur Autonomous Region were enrolled from 2017 to 2019. According to dual-energy X-ray bone mineral density (BMD), they were divided into normal bone mass group, bone mass loss group, osteoporosis group, and severe osteoporosis group, with 15 cases in each group. The lateral femoral muscle tissues of the patients were collected during the operation. The mRNA expression and protein level of COL2 were detected with RT-PCR and immunohistochemistry. The correlation between COL2 expression level and osteoporosis was analyzed comprehensively. Results RT-PCR and immunohistological results showed that there was no significant difference in COL2 expression between normal bone mass group and low bone mass group (P>0.05). The expression level of COL2 in bone mass loss group was significantly higher than that in osteoporosis group, and the difference was statistically significant (P<0.05). COL2 expression level in osteoporosis group was significantly higher than that in severe osteoporosis group, and the difference was statistically significant (P<0.05). Conclusion COL2 expression level is negatively correlated with osteoporosis. COL2 may be a therapeutic target for preventing hip fractures in the elderly.
